to study the impact of toxic pollutants on the living organism of the general population, in
contrast to professional pharmacology or toxicology which target limited groups in their
professional and medical milieu with well-known diagnostic and therapeutic approaches.
Indeed, many food biological matrixes are contaminated with pesticides, polycyclic aromatic
hydrocarbons and heavy metals with very little concentrations that we consider such
negligible daily doses but which could accumulate with the time because their solubility in
different organs. This study focuses on the preventive effect of some bioactive compounds
such as vitamin C, vitamin E, quercetin and phenolic extract from orange peel Citrus
Senensis, against the toxic impact of a polycyclic aromatic hydrocarbon, the naphthalene and
endosulfan, as organochlorine pesticide on brain and immune systems. The results showed a
significant change in neuronal redox status in cytosol and brain mitochondria. Indeed,
endosulfan and naphthalene administered alone or in mixture at the respective doses of 2mg /
kg and 50 mg / kg, showed a disturbance in the balance prooxidant / antioxidant in cell
compartmentalization since they caused a significant fall in the level of GSH, GST, SOD and
CAT cytosolic rats treated with these chemicals. The results of these studies have also shown
a significant increase in MDA cytosolic and a highly significant reduction of cytosolic
proteins level was also recorded compared to the control group. Furthermore, the results of
this work have showed a very significant decrease in mitochondrial enzymatic activity of
SOD and CAT parallel to fall of GSH rate. Increased peroxidation was recorded via the
significant increase in mitochondrial MDA following animal exposure to endosulfan alone at
a dose of 2 mg / kg, reflecting a loss of integrity mitochondrial membranes. Conversely, the
administration of quercetin (10 mg / kg) associated with endosulfan (2mg / kg) was able to
prevent this neuro-mitochondrial toxicity in rats. The in vitro study of the mitochondrial
integrity showed, in the presence of increasing concentrations of endosulfan, a decrease in
absorbance of mitochondrial suspensions, demonstrating that mitochondria were received
massive internalization of water via the opening giant mitochondrial pores (MPTP), causing
them to swell. Inversely, the tubes containing endosulfan and quercetin showed similar
absorbance to the control tube, which provides information on the maintenance of intact
mitochondrial integrity which not allowed undergoing this swelling.
This study also showed a toxic effect on the immune and blood systems through a very
significant increase in the overall number of lymphocytic cells in rats of groups treated with
the mixture endosulfan and naphthalene. It was also found that in the same study, a significant
and very significant respective reduction in granulocytes and monocytes rates. The evaluation
of some blood biochemical parameters did appear impairment of liver function, kidney and
pancreas via the significant increase in transaminases enzymes in the blood serum (TGO and
TGP), cholesterol, triglycerides, urea, creatinin and blood glucose compared to the control
group. Meanwhile, bioactive substances used in these studies, warned the occurrence of these
disturbances in the function of these organs in animals exposed to these persistent organic
products. In conclusion, the results of this present study showed that endosulfan and
naphthalene have neurotoxic power and immunomodulator effect in environmental doses
which have been administered individually or associated in rats. Moreover, it appears that

Index. décimale : 570 Sciences de la vie. Biologie Résumé : Actinobacteria are a group of filamentous bacteria, mainly inhabit the soil. They are known for producing a wide variety of bioactive molecules, especially antibiotics. Is in this context that our research work focused. A total of 99 isolates were recovered from a semi-arid soi,collected from; rhizospheric associated soil (Khenchela), mountain soil (Mount chélia region, khenchela) and Sebkha soil (Oum El bouaghi). Three strains (SF10, SFKH4 and SFKH10), were selected, based on preliminary screening for antimicrobial activity. The isolates were identified as belonging to the genus Streptomyces, based on phenotypic and chemotaxonomic study. Phylogenetic analysis revealed that the strains SF10, SFKH4 and SFKH10 were most closely related to Streptomyces youssoufiensis, Streptomyces spororaveus and Streptomyces coeruleorubidus with a pairwise 16S rRNA gene sequence similarity of 99, 8, 99 % and 99%. Ethyl acetate extracts provied by the culture of the strains on solid culture were tested for their antimicrobial activity against the test microorganisms. The structure elucidation of the biologically active molecules, purified by different chromatographic methods, was carried out by coupling several techniques of mass
spectrometry and Nuclear Magnetic Resonance (1H NMR, 13C NMR, MS, MS tandem, IR and UV-Visible). Streptomyces youssoufiensis SF10 strain produced several molecules belonging to ionophoric polyethers family, abierixin, nigericin, epinigericin, and the newly isolated grisorixin methyl ester. Moreover, only one molecule, belonging to macrotetrolides family (dinactin), was obtained from Streptomyces coeruleorubidus SFKH10 strain. In addition, the partial characterization of the molecules secreted by S. spororaveus SFKH4 strain has shown its capacity to produce a large variety of complexes compounds having high molecular weight. The presence of epimeric forms of nigericin/epinigericin and grisorixin/epigrisorixin has spurred DFT (Density Functional Theory) computational calculations. This analysis was able to provide the relative stability of the most favored epimers. This consideration is of general interest and can be applied to the wide class of naturally occurring polycyclic polyethers. On the other hand, the study of the cytotoxicity of the purified and identified molecules revealed that both nigericin and grisorixin methyl ester showed to affect glioblastoma stem cells proliferation in a dose-dependent manner, with a higher activity for the more lipophilic grisorixin methyl ester (IC50 values of 3.85 and 3.05 μM for VIPI and COMI humain glioblastoma stem cells, respectively). In conclusion, the present study has revealed the potential of semi-arid Algerian ecosystems as a good source for bioactives molecules-producing actinobacteria, specialy Streptomyces youssoufienisis SF10 strain whose secondary metabolites profile has not been studied so far by bioguided method. The clear antiproliferative activity of grisorixin methyl ester on glioblastoma stem cells represents a promising starting point aimed to the development of new drug candidates in the treatment of glioblastoma multiforme.

The work undertaken in this dissertation focuses on the modeling of a storage tank in order to understand better the thermal behavior of the latter one. The impact of many parameters (water injection debit, the nature and the thickness of the insulation, usage of a heat exchanger or an electrical booster) on the temporal evolution of the temperature (in case of charge and of discharge) in the tank was examined.
The modeling of the stratification is undertaken by subdividing the tank into N (20) layers. An energy and a masse balance is drawn up in every node. The system of the global equations is resolved by the Crank-Nicolson method.
The results obtained show clearly that the last layer heats faster, especially as the injection debit is elevated and the tank homogenization time becomes shorter.
Besides, it appears that a good isolation allows to minimize the losses, and to avoid the creation of convective current, which could degrade the stratification.
