In this thesis, aerial parts of two plants belong to Asteraceae family were studied. The first is Echinops spinosus, species distributed around the world and is commonly used in the treatment of various diseases. The second one is Pulicaria undulata, species present in desert areas of North Africa and all of the Middle East regions and mainly used in the treatment of various diseases.
For experimental purposes, the Echinops spinosus sample plants were collected from the
north-eastern part of Algeria in a region near Annaba, whereas the Pulicaria undulate, was harvested in the Djanet region (Tassili ) in the southeastern part of Algeria.
During the laboratory work, different extraction and separation methods were used as
maceration, chromatographic column, TLC and HPLC preparations. Additionally, and in
order to determine the structures of the isolated compounds, deferent spectroscopic methods were combined: nuclear magnetic resonance and its different applications (1H-NMR, 13C NMR, DEPT, COSY, HSQC, HMBC, NOESY and NOE difference) as well as spectrometry of mass high resolution, UV, IR and chiroptical spectroscopy (Polarimetry, Electronic Circular Dichroïsm ECD).
Seven phenolics compounds from Echinops spinosus were isolated and identified: One
aglycone flavonoid, five glycosids flavonoids and one quinic acid. For the first time, ten sesquiterpenes were obtained from the chloroformic extract of Pulicaria undulata (nine sesquiterpenes lactones belong to the Asteriscunolide type and one humulene)
The biological study includes the antioxidant activity of CHCl3, AcOEt and n-BuOH extracts for Echnops spinosus L.and the anti-inflammatory activity of the CHCl3 extract for Pulicaria undulata L., and that of two isolated compounds.
Analysis GC and GC/MS of the essential oil of Pulicaria undulata L. identified 31
compounds. The main components are carvotanacetone (14,8%), δ-cadinene (8,2%),
α-cadinol (4,7%), epi-α-cadinol (3,4%) and thujanol (4,7%).

Index. décimale : 540 Chimie et sciences connexes Résumé : Although Senecio species are known as sources of potentially toxic pyrrolizidine alkaloids (PAs), some species of this genus are traditionally used as remedies. In this work the evaluation of biological activities and the analysis of PAs of Algerian specimens of Senecio delphinifolius Vahl (its vernacular name in Algeria is ""Hachichetettolba"") are reported. Thus it was possible to identify six APs by GC and GC-MS analysis: (1) senecionine (2) seneciphylline (3) integerrimine (4) senkirkine (5) dehydrosenkirkine and (6) neosenkirkine, this technique is frequently used for complex mixtures of such compounds. The species S.
delphinifolius, contains 140 ppm of PAs belong to the toxic series (1, 2-unsaturation in the pyrrolizidine cycle and macrocyclic diester), the use of S. delphinifolius should be discouraged in traditional medicine.
Our research has also led to the isolation by conventional purification techniques of three compounds from the n-butanol extract of the specie S. delphinifolius Vahl two of them are glycosylated flavonols, (7) Quercetine 3-O-α-(rhamnopyrannosyl (1-6) glucopyrannose) [rutine], (8) Kaempferol 3,7-di-O-α-Lrhamnopyranoside [Kaempferitin], the third is a derivative of 1, 4-benzoquinone that are common in the
genus Senecio (9) 2, 5 Cyclohexadiene-1-acetic acid, 1-methyl-4-oxo, methyl ester. A fourth product was isolated from the dichloromethane extract, the structure is being identified. The structures of the isolated compounds were elucidated through the use of NMR techniques 1D and 2D, mass spectroscopy, UV spectroscopy and acid hydrolysis. (10) Chlorogenic acid was identified by co-chromatography with standards.

Index. décimale : 540 Chimie et sciences connexes Résumé : Our phytochemical and biological works concerned the aerial parts of two species belonging to Asteraeae family : Phagnalon saxatile (L) Cass. and Phagnalon sordidum L.
Several chromatographic methods allowed us to isolate ten products from the n-butanol and ethyl acetate extracts of Phagnalon saxatile from which one is new, and twenty one products from Phagnalon sordidum.
Structures were determined by different spectral methods: nuclear magnetic resonance with different applications (NMR 1H, NMR 13C, COSY, HSQC and HMBC), the mass spectrometry, UV, acid hydrolysis as well as comparison with literature data.
4 methods were used to evaluate the antioxidant activity of Phagnalon sordidum and Phagnalon saxatile. The latter showed a good activity by the use of 4 methods: DPPH, ABTS, CUPRAC and superoxide radical scavenging activity by alkaline DMSO whereas Phagnalon sordidum exhibited a moderate activity with DPPH and CUPRAC tests.
Furthermore, petroleum ether and chloroform extracts from Phagnalon saxatile showed moderate antibacterial activity against the tested strains. The isolated products (1-O- β-D-glucopyranosyl-1,4-dihydroxy-2-((E) 2-oxo-3-butenyl) benzene, 1-O-β-glucopyranosyl- 2(3’,3’-diméthylallyl) hydroquinone and Potassium 4-hydroxy-3-methoxybenzoique acide
methyl ester-5-sulfate) from Phagnalon saxatile showed a moderate cytotoxic activity by the use of MTT method.
