Silybum marianum or milk thistle is a medicinal plant, belonging to the asteraceae family, the
most widely used throughout the world, for its richness in bioactive substances. They have
been evaluated, in vitro, for their antioxidant activities and their total polyphenol and
flavonoid content. For the present study, four extracts (ethanolic (EE), butanol (EB), ethyl
acetate (EAE) and dichloromethane (ED) were prepared from the aerial part of the medicinal
plant Silybum marianum. Antioxidant power was achieved using the free radical scavenging
tests, DPPH and ABTS, and the CUPRAC test. Total polyphenols and flavonoids were
determined by spectrophotometric methods.
The results revealed that the extraction yields vary depending on the solvent used, they are
between 12,17% and 20,5%. The IC50 vary from 576,82 ± 3,43 μg / mL to 42,5 ± 1,51 μg /
mL with DPPH, of which the ethyl acetate extract of Silybum marianum was the most active
(IC50 = 42,50 ± 1,51 μg / mL) than other extracts. For the test with ABTS, the IC50 are
between 142,67 ± 1,85 μg / mL and 12,56 ± 1,19 μg / mL, and the dichloromethane extract
was the best scavenger of this radical with an IC50 of 12 , 56 ± 1,19μg /mL. Regarding the
CUPRAC test, the extracts showed an activity of between 31,25 ± 1,38μg / mL and 437,78 ±
5,36 μg / mL, where the ethyl acetate and dichloromethane extract of A0,5 the lowest (31,75 ±
0,45 and 31,25 ± 1,38 μg / mL, respectively), and therefore, the most important antioxidant
activities. The quantification of total phenols by the Folin-Ciocalteu method and of flavonoids
by the AlCl3 method gave high values in the ethyl acetate extract, where the highest value is
estimated by: 266,45 ± 3,88 mg / g and 229,18 ± 5,91 mg / g. These results prove that Silybum
marianum is very rich in different phenolic compounds and exhibits good antioxidant activity.
It could be used in the pharmaceutical field as a potential source of bioactive molecules with
antioxidant properties. |
