In this thesis, we first synthesized a series of nitrogenous organic compounds classified into four families which are oximes, azomethines, phenylhydrazones and semicarbazones. We subjected them to antioxidant activity tests (DPPH, CUPRAC and iron reducing power). Moreover, molecular modeling calculations with density functional theory (DFT) have been carried out on our molecules in order to determine certain molecular descriptors (BDE, IP, PDE, PA and ETE) which allow us to analyze and to explain the experimental results. All calculations were performed using the Gaussian 09 software. Finally, we tested their anti-enzymatic activity and performed in parallel, it’s in silico simulation through a molecular docking calculation. The antioxidant activity tests were very satisfactory and showed a well-established antioxidant power for the tested compounds, especially for the DPPH scavenging test and the CUPRAC test. Indeed, for these two tests some molecules had a much better activity than the standard antioxidants used: butylated hydroxyanisole (BHA) and butylated hydroxytoluene (BHT). On the other hand, the values of the molecular descriptors obtained are in good agreement with the experimental results. The analysis of these descriptors allowed us to propose mechanisms for the radical activity of our molecules. On the other hand, the tests of the anti-enzymatic activity revealed a moderate activity for the tested compounds and the enzyme-ligand interactions thus engaged, are listed after the analysis of the results of the molecular docking carried out. The results obtained showed a good correlation between the theoretical and experimental studies, confirming the antioxidant power of our compounds, which could allow them to be used in the pharmaceutical and food industries, while noting their obedience to Lipinski's rules. Finally, further tests are recommended to ratify these results.

Our phytochemical and biological work concerns the aerial parts of two endemic Saharan species belonging to the Fabaceae family: Ononis angustissima Lam. Var and Lotus corniculatus L. Several chromatographic methods were used in our experimental work which allowed us to isolate nineteen products from the butanolic extract of Ononis angustissima Lam. Var. from which two are new, and eight products from Lotus corniculatus L. The structures were established by different spectral methods: nuclear magnetic resonance with different applications (NMR 1H, NMR 13C, DEPT, COSY, HMQC and HMBC), the mass spectrometry, UV, as well as comparison with literature data. The antiproliferative activity tests of the n-butanol extract and the compounds isolated from L. corniculatus were carried out on three cell lines: J774.A1 (murine macrophages monocytes), WEHI-164 (murine fibrosarcoma) and HEK-293 (human epithelial renal cells) have shown that the n-butanol extract and the pharbitoside A compound are the most active against the three cell lines. The two extracts and most of the isolated compounds of Ononis angustissima Lam. Var have excellent activity, so these molecules are considered first class antioxidants compared to reference molecules. They can therefore be used for therapeutic applications. Finally, the quantitative estimate of total polyphenols has shown that these extracts have a high content of polyphenols.Note de contenu :

of the septentrional Sahara from the Cistaceae family: Helianthemum ruficomum and
Helianthemum confertum. To our best knowledge, no studies are reported so far for these
species. The purpose of this work was to investigate the chemical composition and the
radical scavenging capacity of these species and their isolated components.
The ethyl acetate and «-butanol soluble parts of the aqueous-ethanol extracts of the
aerial parts of these species were evaluated for their free radical scavenging capacity by
ABTS'+ assay. The obtained data which showed important antioxidant powers were
confirmed by TEAC and ORAC assays. On-line HPLC- ABTS'+ assays were then applied to
screen the extracts. The results which revealed remarcable wealth in antioxidants allowed
guiding the fractionation of the extracts by column chromatography on silica gel 60. The
obtained fractions were purified by chromatography on preparative plates of silica
gel 60 and/or by semi-preparative reverse phase HPLC leading to the isolation and the
identification of 14 compounds from H. ruficomum and 6 compounds from H confertum. The
compounds were identified by spectral analyses, mainly ESI-HRMS (+), UV, ID NMR (*H,
13C, DEPT) and 2D NMR (DOSY, COSY, NOESY, HSQC and HMBC) and comparison of
their spectroscopic data with those reported in the literature.
The antioxidant power of the isolated compounds was also evaluated by on-line
HPLC-ABTS'+, TEAC and ORAC. The results clearly indicate high antioxidant potential
of the extracts and tested compounds isolated from these two species.

They are Centaurea choulettiana Pomel and Centaurea fragilis Dur.
The phytochemical investigation of cyclohexane, ethyl acetate and n-butanol extracts from the aerial parts of C. choulettiana and n-butanol extract of C. fragilis allowed the isolation and identification of 13 products. The structural elucidation of the isolated compounds was carried out by combining the various spectroscopic methods (UV, 1D & 2D NMR, HPLC/TOF-MS and GC/MS) as well as by comparing the results with the data of the literature.
The analysis of essential oil (flowers) of C. choulettiana by the GC/MS technique, carried out for the first time, showed the presence of several chemical groups from which, phthalate (18.38%) followed by β-eudesmol (6.8%), 5-epoxysalvial-4(14)-ene (6.6%), caryophyllene oxide (4.28%), tricosane (3.31%), 7-epi-α-eudesmol (3.08%) and heptacosane. The antibacterial activity of this oil was determined against five bacterial strains according to the disk diffusion method, the results showed modest activity against all the strains.
Quantification of the total polyphenols and total flavonoids of the EtOAc and n-BuOH (leaves and flowers) extracts of C. choulettiana was performed by colorimetric methods. This step showed a content of polyphenols and flavonoids more important in the AcOEt extract of the leaves. In order to identify and quantify the phenolic acids present in these extracts, chemical screening was performed by HPLC-TOF/MS, this revealed the presence of 9 phenolic acids in various amounts. The HPLC-TOF / MS analysis of the n-BuOH (leaves) extract of C. fragilis revealed the presence of 9 phenolic acids and 10 flavonoïds.
The antioxidant activity of the EtOAc and n-BuOH (leaves and flowers) extracts of C.
choulettiana was evaluated by the free radical scavenging method (DPPH) and the inhibition of lipid peroxidation (LPO). The EtOAc (leaves) extract exhibited the strongest effect with respect to the DPPH free radical (96.54%) and the inhibition of lipid peroxidation (64.17%).
Concerning the antiproliferative activity, the results showed that the cyclohexane extract of C.
choulettiana was the most active against HeLa cells (carcinoma of the human cervix), while the n-BuOH extract of C. fragilis was inactive in his unlevel doses.

Biologie Résumé : The medicinal plants represent an immense source of bioactive molecules, endowed with numerous biological activities. The present work concerns of photochemistry study of flavonoids isolated from Thymus serpyllum, Thymus algeriensis, and Salvia officinalis, and an evaluation of their antioxidant and antibacterial activities.
The rate of total phenols ranged from 16, 64 to 25,47 mg GAE / g extract, whereas the rate of flavonoids is 1,7 to 2,4 mg QE/ g extract. The antioxidant activity is seems very important, whose the results vary from 88% to 92% for T. algeriensis, 81% to 92% for T .serpyllum, and 87% to 92% for S. officinalis.
