Biologie Résumé : Hyperhomocysteinemia (HHcy), oxidative stress and decreased antioxidant capacities lead to several clinical manifestations and particularly, cardiovascular and liver diseases. In this study, we evaluated the protective and the preventive effect of the crude extract of Argania spinosa against HHcy ,hyperlipidemia, oxidant status and damages in the aorta, heart and liver induced by high L-methionine intake in mice. After 3 weeks of treatments, Hcy concentrations, lipid parameters, liver enzyme activities, hepatic antioxidant status and histological sections of aorta, liver and heart were determined. Our results showed that consumption of high L-methionine diet (500mg/Kg)
led to an increase in plasma Hcy, CHO, LDL-c, TG, AST, corresponding with decrease of HDL-c, reduced GSH, and catalase activity. These were associated with the loss and degeneration of endothelium, fenestration and formation of foam cells in the media of the aorta, also the alteration of the cardiac muscle and liver tissue. However, the administration of the crude extract of A. spinosa (150mg/Kg) in combination with L-methionine ameliorated all these changes.
Moreover, in the present thesis, we conducted a study in vitro to evaluate the antibacterial effect of Argan oil on biofilm formation. The results showed that there was a trend of increasing inhibition of 4 species of bacteria (Streptococcus mutans, Streptococcus anginosus, Streptococcus intermedius, and staphylococcus haemolyticus), that belong to gram positive strain as the Argan oil was getting more concentrated.

There is a balance in the metabolism of Hcy at physiological conditions.
At high level, the hyperhomocysteinemia (HHcy), considered as an independent risk factor in atherosclerosis, which can cause cellular and molecular alterations.
In the present study, we have measured thiol amino acid (homocysteine), lipidic statut (total cholesterol, LDL, HDL, triglycerides), and activity of transaminases (GOT, GPT) in mice administered with (400mg/kg) of L-methionine during 21 days. Following oral administration of L-methionine in high dose, its degradation product Hcy which is markedly
elevated in plasma and exerts an angiotoxic action directed to the iliac aorta and the other organs (liver, heart and kidneys). This is showed degradation and desquamation of endothelial cells also formation of foam cells in the media part with oval nuclei in mediocytes.
However, the aortic intima of the groups of mice that had been fed with L-methionine plus S.
mialhesi extract (500mg/kg) and also with vitamins B9 and B12 (0,7mg/kg and 0,75μg/kg), respectively showed intact endothelium and spindle-shaped mediocytes nuclei. Nevertheless, oval-shaped mediocytes nuclei and a few foam cells were observed. In the histological sections of the cardiac muscle, liver and kidneys no lysis was observed.
The vitamins B9 and B12 serve as cofactors in the metabolism of L-methionine. So, the vitamins therapy and the extract of S. mialhesi in mice have reduced Hcy, total cholesterol, triglycerides and LDL however, the concentration of HDL was increased.
The extract of medicinal plant S. mialhesi used in this study has antioxidant effect as the vitamins B9 and B12 in mice.

Biologie Résumé : Angiogenesis is the formation of new blood vessels from pre-existing vessels, existing in many pathophysiological conditions, including tumorogenesis. In the activation of the endothelial cell (EC),VEGF-A (vascular endothelial growth factor) is the decisive stage of the process. Since homocysteine (Hcy) has an angiotoxic power, the aim of this work is to assess (in vivo and in vitro studies) the effect of this molecule on angiogenesis and tumor invasion involving VEGF and its two powerful signaling effectors: the HIF(hypoxia –inducible factor )
-1α and the OPN (Osteopontin).
In the in vivo study, the conception of our Hyperhomocysteinemia model (HH) is
confirmed by biochemical study (total Hcy, total protein, CRP and MDA) and histological one (aorta, liver and thymus). We showed that HH potentiated the presence of VEGF-A in thymus, suggesting the installation of a reparative angiogenesis effect, in the same time to an inhibition of VEGF-A and HIF-1α expressions in aortic tissue.
In the in vitro study, we evaluated the antiangiogenic effect of Hcy previously
revealed in vivo, by exposing HUVECs to cytotoxic and pathophysiological concentrations of Hcy. In fact, the first range revealed a strong inhibition of the proliferation, migration and capillary sprouting associated with senescence and apoptosis of EC. In pathophysiological range, inhibition is always dose -dependent and became completely significant in adenosine condition. Hcy seems to act by an hypomethylation mechanism. In addition, the results of qRT-PCR revealed that Hcy inhibited gene expression of VEGF-A and OPN in dosedependent manner.
This study shows a new aspect of antiangiogenic properties of Hcy, by impaired the

Biologie Résumé : Neural tube defects (NTDs) represent a major cause of infant mortality among congenital malformations, whose pathogenesis remains poorly understood. In this study realized on an algerian population of affected children and their mothers, we investigated the frequency of many polymorphisms involved in the of folates / homocysteine metabolisms, Their possible contribution in the etiology of NTDs, as well as their influence on folates and homocysteine concentrations.
The mutations were determined by the PCR / RFLP method, the assay of the biochemical parameters was performed by chemiluminescence.
The analysis of genetic polymorphisms has shown that AG and AA variants of the G80A polymorphism of the RFC gene are associated with the increased risk in affected children.
Moreover, we have observed that the combined effect of C677T and A1298C polymorphisms in MTHFR gene, confers a risk for the occurrence of these abnormalities in our population .Indeed, statistical test of interaction between the two polymorphisms have shown that the combination of heterozygous genotypes(MTHFR 677CT/1298AC) elevate the risk in NTD cases (p <0.05).
These two polymorphisms ,in MTHFR gene, affects homocysteine metabolism in mothers of NTD cases leading to homocysteine concentration values higher in mothers with TT genotype of the C677T (p < 0.05) and that of AA genotype for the polymorphism A1298C (p <0.05).
For the polymorphism A2756G of the MTR gene, Although, Its association with NTDs risk appears to be negative, It was found to decrease the RBC folate level strengthen the contribution of A2756G mutation in NTDs occurrence (p <0.05).
Data on the association between MTRR A66G, CBS 844ins68 polymorphisms and plasma homocysteine and serum/RBC folate concentrations as well as the relationship between this polymorphisms and NTDs risk are inconclusive.
This original study by its primacy in Algeria and also in North Africa, both in terms of the diversity of polymorphisms examined and the nature of the population (children affected) deserves, encourages the extension of the study to a larger sample In order to show the interactive effect of these mutations on the occurrence of NTDs and on the rate of metabolites.
