This thesis sheds the light on the action of these antioxidants on the oxidative stability of a whey-based spread-type emulsion, but also the crystallization of fats in the fatty phase. The crude phenolic extracts of Pituranthos scoparius and Hibiscus sabdariffa and the essential oils of the aerial parts of P. scoparius were studied. These extracts were obtained via conventional extraction techniques (decoction and hydrodistillation) and non-conventional techniques (microwaves and ultrasounds). Spectrophotometric assays were performed for the assessment of the contents of total phenolics in the crude extracts studied. Various chromatographic
techniques have been performed for the characterization and identification of the different classes of antioxidants. The antioxidant activity was studied by the tests of reducing power and antiradical activity. Different emulsions were formulated at both laboratory and pilot scales, incorporated by the concentrated crude phenolic extracts of the plants and the essential oil from stems of P. scoparius at different concentrations. Their oxidative stability was evaluated by Rancimat test. The two phases of the emulsion have been studied regarding two aspects: whey protein-polyphenol interactions in the aqueous phase and lipid crystallization in the fatty phase containing palm, soybean and sunflower oils. A Tricine SDS-PAGE was performed in the former and a set of investigation techniques for the study of isothermal crystallization (DSC, p-NMR and PLM) in the latter were applied. The results of spectrophotometric, chromatographic and antioxidant activity corroborate with those of oxidative stability and showed a protective effect against lipid oxidation at low concentrations of plant crude extracts (100 and 300 ppm) and the essential oil of P. scoparius (100 ppm). The sensory evaluation of the spread at 100, 300 and 600 ppm of the essential oil of P. scoparius, with a principal component analysis (PCA), revealed the influence of some sensory attributes on the segregation of emulsions at these different concentrations. The interactions of caseins and β-Lactoglobulin with polyphenols in the aqueous phase, with the formation of characteristic dimers, were revealed by densitometry and LC-MS/MS analysis. The formation of these dimers did not affect in any case the oxidative stability of the emulsions tested. The study of the effect of essential oils of stems and flowers of P. scoparius on lipid crystallization (palm oil alone and a mixture of the oils) showed a change in isothermal crystallization behavior of triglycerides with a delay of crystallization, a decrease in calculated enthalpies of fusion, slower crystallization rates and a drastic decrease in solid fat contents (SFC). This modification is also noticeable and different shapes and sizes of triglyceride crystals (spherulites) were visualized for fats with essential oils compared to control ones. The induction of a dilution effect of the system by the essential oils is discarded and the hypothesis of the implication of major monoterpenes in disturbing the crystallization events (nucleation and crystal growth) is proposed. From this study, it can be concluded that the recovery and industrial exploitation of the two plants studied is strongly encouraged, especially for P. scoparius which is already exploited in the Algerian traditional pharmacopoeia and food preservation. In addition to the antioxidant effects reported for the essential oil, it is suggested the possibility of its use as a natural additive to modify fat crystallization for applications in shortenings and various food emulsions.

This work is devoted to the phytochemical, biological and pharmacological study of two endemic Saharan species; Helianthemum lippii (Cistaceae) and Cymbopogon schoenanthus (Poaceae). The extraction of the two species followed by the chromatographic separations (CC, CCM and HPLC semi-preparative), led to isolate and identify using spectroscopic methods (1D and 2D NMR) 6 compounds among which two sterols, as well as four sesquiterpenes: Stigmasterol, β- Sitosterol, (1S,4aa)-4Methylene-6-(1-hydroxy-&-methyethyl)-8a-b-methyl-decahydroxydronaphtalene-Ab-ol 1S, 7S, 10S, eudesm-4(14)-en-1α-11α-diol(hydroxy-β-eudesmol), 4α, 11-dihydroxy-7-epimereudesmandiol (7-epi-cryptomeridiol), 4β, 7β, 11-enantioeudesmantriol (Pterodontriol), 4β,6β,10α-trihydroxy guaiane sesquiterpène (Teuclatriol). Chromatographic analysis of the essential oils of Cymbopogon schoenanthus showed that the hydrocarbons represented 26.7% while the sesquiterpene fraction reached 40%. The HPLC-TOF / MS analysis of the AcOEt and n-BuOH extracts of the two species suggested the presence of 32 phenolic acids and flavonoids which were present in different proportions The efficacy of inhibiting DNA damage and the evaluation of the anticancer activity carried out on the EtOAc and n-BuOH extracts of the two species demonstrated a particularity observed in Helianthemum lippii on the antioxidant effect and thus the antiproliferative effect. The pharmacological study was carried out on the effect of the extracts on the distal colon of Wistar rats; which have shown a marked relaxing effect on the isolated distal colon of the rat which could represent an underlying mechanism for the traditional use of these plants for gastrointestinal conditions. Further studies will help us understand the metabolites responsible for the effect and mechanism involved.

ICP-M" Index. décimale : 540 Chimie et sciences connexes Résumé : Our goal in this work was to search the secondary metabolites of flavonoids type from two species from Asteraceae and Cistaceae families in order to search new natural compounds of biological interest. Thirteen (13) extracts from different species including twelve (12) from the Aures regions were subjected to a phytochemical screening. On the basis of these preliminary results, the phytochemical investigation of the aerial parts two species, Pulicaria inuloides and Cistus albidus from the Bechar and the Aures regions respectively was carried out. This approach allowed the isolation of twelve natural compounds, among which eleven flavonoids, one phenolic compound as well as two acetyl derivatives of one of the isolated compounds were characterized using spectroscopic methods such as UV-Visible, IR and NMR.
The biological activities in vitro (antioxidant and antibacterial activity) as well as the total phenolic and flavonoids compounds of the various extracts were discussed. We also reported the evaluation of the in vitro antiprotozoal activity of the two species Pulicaria inuloides and Cistus albidus against Leishmania amazonensis, Trypanosoma cruzi and Acanthamoeba castellanii str. Neff. The bioguided fractionation of the CHCl3 extract from the aerial part of P. inuloides leads to the isolation and characterization of quercetagetin-3,5,7,3'-tetramethylether as the active ingredient. Two acetylated derivatives were prepared from this natural compound and these three compounds were evaluated for their activities against the promastigote form of L. amazonensis.
All these compounds exhibited moderate leishmanicidal activity with IC50 values ranging from 0.234 to 0.484 mM and low toxicity to macrophage cells (CC50 ranging from 0.365 to 0.664 mM). This study reported the antiprotozoal evaluation of Pulicaria inuloides for the first time and the results show that this species can be considered as a source of anti-leishmanicidal agents. The bioguided fractionation of the active extract of CHCl3 from C. albidus against T. cruzi led to the isolation of two fractions F6,4 and F6,5 which showed also a potent activity up to 100 μg/ml. The bioguided fractionation of the active extract of AcOEt from the aerial part of C. albidus against A. castellanii led to the isolation of a fraction F14A which showed a significant activity up to 100 μg / ml. Fractions F6,4 ; F6,5 and F14A consists of product mixtures, which have not been separated.
We have also described the chemical compositions of the essential oils of Pulicaria inuloides and Cistus albidus. The major compounds of the species C. albidus are manool and 15,16-dinorlabd-8(20)- en-13-one whose skeleton of type labdane is a specific marker in this genus, these compounds are not described in samples from other regions and in particular in the Mediterranean regions (Spain and France) that appeared in the literature. The major compounds of the species Pulicaria inuloides are: δ- cadinene, α-muurolene, γ-cadinene, α-epi-cadinol and α-cadinol are not present in the other samples of Pulicaria inuloides from other areas published in the literature.
In this study, the mineral content (inorganic elements) of 21 isolated extracts of various aromatic species of the Sahara (P. inuloides) and the Aures region (Euphorbia helioscopia L., Peganum harmala L., Daphne gnidium L., Calendula algeriensis L., Bunium incrassatum L., Echinops ritro L. Marrubium deserti de Noè., Juniperus oxycedrus L., Juniperus phoenicea L., Marrubium vulgare L., Cedrus atlantica (Endl.) Manetti and Thymus algeriensis L.) were also determined by inductively coupled plasma atomic emission spectroscopy (ICP-AES) and inductively coupled plasma mass spectroscopy (ICP-MS) . The results obtained showed higher concentrations of Ca, K, Mg, Na and S
(mg/kg) in the different plant extracts.

impacts and the social-economic stakes. Therefore, the main encouraging way envisaged to
reduce enteric methane is the use of additives containing plants secondary metabolites.
The objective of this thesis work was to test at first the in vitro potential of essential
oil extracts and condensed tannins-rich plants to reduce methane production. Secondly, an in
vivo study was conducted for a validation of results obtained in vitro and for a better
knowledge of ruminal methanogenesis and fermentative phenomena.
Our work allows concluding that essential oil and condensed tannins have the potential
to decrease in vitro methane production. Essential oils of Mentha pulegium and Juniperus
phoenicea negatively affect in vitro ruminal fermentation. Condensed tannins reduce in vivo
methane production without notable unwanted effects on ruminal fermentation and on
digestibility. The effect of condensed tannins is rather linked to the dose and not to the nature
of the plant. This work also highlighted the absence of relation between concentration and
diversity of methanogens and methanogenesis. Reduction of methane is due to the inhibition
of the activity of methanogens rather than to the decrease of their concentration. The
presentation of tannins rich plants under granular shape had also accelerated the transit.
Results from the in vivo study suggests that dietary condensed tannins from Leucaena
leucocephala, Manihot esculenta, and to a lesser extent from Gliricidia sepium in pellet form
could be used to mitigate methane production with no marked detrimental effects on other

C.mreboudianum and C. fuscatum shows the presence of essential oils, flavonoids, sterols or terpenes,saponins and carotenoids.
The GC/MS analysis of the essential oils of species: Oa, Cƒ and Cr showed that these oils contain major compounds with oxygenic functions (aldehydes, ketones, esters and acids) and that for theathreeaplants.
This study also allowed the isolate, the purify and the identify of three compounds from the flowers of Oa (B-sitosterol, Luteolin-7-O-glucoside and Quercetin-3-O-glucoside), the molecular structures of purified compounds were elucidated by UV spectroscopy techniques and NMR (1H, 13C).
Spectrometric quantitative determinations of total polyphenols by Folin-Ciocalteu reagent and flavonoids those prepared by the aluminum chloride AlCl3 revealed rich extracts (AcOEt and BuOH) by flavonoids and phenols contribution extracts (Hexane and CHCl3) of different species tested.
The antiradical potential of essential oils (Oa, Cƒ and Cr) and extracts (Hexane, CHCl3, EtOAc and BuOH) Oa and chrysanthemum species tested was determined by the method of DPPH and the results show that essential oil of Oa is most active by supplying essential oils (Cƒ and Cr). The extract butanol and ethyl acetate of the two species (Oa and Cm) in addition to the chloroform extract of the species Cƒ, demonstrated a remarkable antioxidant activity.
The antimicrobial potency testing, CMI and CMD has been made on a range of (bacteria and fungi) to give an idea of the scope of our antimicrobial activity (essential oils and flavonoid extracts), the EO of Oa seems to enjoy a remarkable inhibitory activity against Gram (+) tested and Candida albicans compared with the EO of Cƒ and EO of Cr appears less active.
