Biologie Résumé : The present study aimed to evaluate the protective effect of argan oil and selenium against oxidative stress induced by mercury. For this reason, a phytochemical study of antioxidant tested in vitro was performed on argan oil to estimate its expected effect. The results showed the presence of phenolic compounds in a very high concentration. The evaluation of power scavenger argan oil is more active with IC50 = 13.33 ± 1.3, and in vivo on Albino Wistar rats were treated with mercury alone or combined with argan oil and selenium, for a period of three weeks, the blood is then withdrawn and the rats are sacrificed the various parameters are determined. From the analysis of our results, we observed a significant reduction in weight gain during treatment with mercury alone or combined with argan oil and selenium, but no clear change in the relative weight of liver, kidney or spleen was obtained. The results also showed significant changes in biochemical parameters, immunological and haematological. Characterized primarily by a significant increase in serum glucose, urea, creatinine, uric acid, total and direct bilirubin, total lipids, cholesterol, triglyceride, conjugated diene and the enzymatic activity of AST, ALT, LDH and PAL .dropoff window Whereas serum concentrations of albumin and total protein were reduced in treated rats compared to mercury control animals. Results also show immunological and haematological toxicity especially among the treated groups the pure mercury, an increase in serum IL-1, IL-6 and TNF and elevated white blood cell count, decreased red blood cell levels, hemoglobin concentration the percentage of hematocrit and phagocytic activity during treatment with mercury. Thus the treatment of the rats with the mercury causes a reduction in the plasma antioxidant capacity characterized by decrease in the serum concentration of vitamin E and C and rate of DPPH and thiols. We also observed that the results obtained highlight the detoxifier potential expressed by the decrease in erythrocyte glutathione levels, liver and kidney, metallothionein, the enzymatic activity of the antioxidant system SOD, GPx, GST and catalase and increased in lipid peroxidation expressed by the high levels of plasma MDA, erythrocyte, liver and kidney. Also the results clearly showed substantive alterations of tissue studies in comparison with the control, with hepatocyte cytolysis, hepatic necrosis and tubular degeneration with necrosis of the kidneys. Furthermore supplementation argan oil and selenium changed the toxic effects of mercury and decreased histological alterations and showed that both antioxidants are effective against oxidative stress induced by mercury

The haemagglutinating activity of Astragalus armatus was 1:9(512). And the Ocimum basilicum was 1:7(128).Lectins extracts of Astragalus armatus have given a highly selective groupe A erythrocytes. Ocimum basilium extract showed no ability to distinguish human blood groups. For some number of single sugar galactose inhibe the haemagglutinating activity of Ocimum basilicum lectins. The extract Astragalus armatus was stable in the pH range of 3-7,5 and heat stable up to60°C. The extract Ocimum basilicum was stable in the pH range of 2-7,5 and heat stable upto 80°C.The extrct of Atragalus armatus have immune effect. They are often multivalent and have the well-known ability to agglutinate erythrocytes.
Fungi, including microscopic fungi and yeasts, have been the subject of considerable interest in recent years because of their potential biomedical and biotechnological applications. The information on fungal lectins, especially the structural point of view, are minimal compared to those of their plant and animal counterparts. It is in this context that lies our purpose was realized by the biochemical and structural characterization of fungal lectin Terfezia boudieri ( Algerian Sahara Truffle) on which we previously had no such data.
Two lectins: TrFBL1 and TrFBL2 were extracted, purified, characterized and sequened. The structures of 3D TrFBL1 native protein and its complex with a ligand, fucoside sélénométhyl were obtained and are being refined .The structures of TrFBL2 complexes with N-acetylglucosamine and two other glycans were obtained at high resolution.
The Genome of Terfezia boudieri was obtained and is available from the month of December 2013 and will become part of the database collected Lectin 3D (http://lectin3d.cermav.cnrs.fr).

Biologie Résumé : Lectins are ubiquitous proteins, non-immune origin, presenting several biological
properties, due to their unique ability to read the biological information codified in the threedimensional structure of sugars and glycoproteins. In this work, 6 strains of actinomycetes, which 5 belonging to rare genus were tested for their production of lectins. The hemagglutination test was used to select the strain Micromonospora aurantiaca GF 44c as best producing strain of these molecules. The study of the lectins contained in the intracellular fraction of strain Micromonospora aurantiaca GF44c led to the discovery of membrane vesicles of nanometric size similar to those found in other Gram-positive bacteria (30 to
100nm) through the use of different technical (FPLC Purification on Superose 6, transmission electron microscopy and dynamic light scattering). These vesicles have the ability to induce, in addition to hemagglutination, immunomodulatory and proteolytic activities. The SDSPAGE shows that these vesicles are very rich in diverse proteins. The proteomic analysis by mass spectrometry of these membrane vesicles identified 81 different proteins, mainly adhesins and membrane transporters (Substrate binding proteins and ABC transporters), enzymes (metalloprotease, peptidase, succinate dehydrogenase, Prolipoprotein diacylglyceryl transferase, DCSA oxidoreductase, Glutamine synthetase, N-acetylmuramoyl-L-alanine amidase), and lipoproteins. Functional analysis of these vesicular protein, suggest that these
vesicles play an important role in the survival of the bacterium in a stressful particular biological environment, by the nutrient acquisition (amino acids carbohydrates and inorganic ions), the resistance to stress factors through the action of several enzymes, and redirect the immune system of the host by a strong immunostimulation. Biologie Résumé : Lectins are present in all living organisms and are used as tools in research and in the
biomedical sector. Currently, the microbial lectins are attracting more and more of interest
because of their diverse biological activities.
Some strains of actinomycetes Rhizospheric promoters of plant growth and antagonistic fungi and pathogens of these plants were chosen to highlight the production of lectins to purify characterize and study their biological activities. The six strains studied were identified to genera Streptomyces and Nocardiopsis. The strains were cultured on two liquids (Bennet and ISP2). The adhesive strength of the extracellular and intracellular extracts of the culture media is evidence placed in the presence of seven types of erythrocytes.
Both extracts of six strains, in any case of the medium, have no agglutinate human erythrocytes.
Strongest agglutination was obtained with the presence of intracellular extracts and rat erythrocytes of rabbits. The ISP2 environment better promotes the production of lectins. Intracellular fractions strongly caking rat erythrocytes were retained. That of Pru16
strain for purification and characterization and that of the Vic8 strain to study the
immunomodulatory effect.
Lectins, the intracellular fraction of strain Pru16 underwent a first primary purification
with ammonium sulphate and gel filtration G75. Differential precipitation showed that the
first three fractions exhibit HA activity. After filtration, two peaks were observed (peaks A
and B). Peak B shows no HA activity. However, the specific activity of the crude extract was
8.37 HU / mg, and it increased to 14.95 UH / mg after ammonium sulfate precipitation, and
20 UH / mg (picA) .after G75 gel filtration. The HA activity of this fraction remains at
alkaline pH and high concentrations of EDTA.
After column chromatography mucin sepharose 4B (second purification), two peaks
are observed. The proteic fraction 33 peak 2 has a high specific HA activity about twelve
times greater than that observed for the crude extract (8.37 111.49 against UH / mg), however
the first peak shows no HA activity. The affinity test conducted with the proteic fraction 33 of Peak 2 showed that the Pru16 strain binds specifically to protein. The SDS-PAGE analysis
showed that the hemagglutinin contained in the portion 33 of the P2 peak is located in the
band 16kDa The identification of proteins 16kDa band, after digestion and elution of peptides by LC, was top by tandem mass spectrometry proteomics comparing with the database related
strains.
The identified analysis of 6 different proteins similar, one of them has a high score,
with a molecular weight similar to that for protein band. This is the lysozyme C who did him
no HA activity. Our protein of 16kDa presents antimicrobial activity.
The enhancement of the phagocytic index of the corrected phagocytic index, and decreased half-life time in circulation of carbon particles in the blood indicate that the Vic8 strain of the crude extract possess one or more factors able to significantly boost the phagocytic activity of macrophages and reticuloendothelial system in mice. Biologie Résumé : Lectins are present in all living organisms and are used as tools in research and in the
biomedical sector. Currently, the microbial lectins are attracting more and more of interest
because of their diverse biological activities.
Some strains of actinomycetes Rhizospheric promoters of plant growth and antagonistic fungi and pathogens of these plants were chosen to highlight the production of lectins to purify characterize and study their biological activities. The six strains studied were identified to genera Streptomyces and Nocardiopsis. The strains were cultured on two liquids (Bennet and ISP2). The adhesive strength of the extracellular and intracellular extracts of the culture media is evidence placed in the presence of seven types of erythrocytes.
Both extracts of six strains, in any case of the medium, have no agglutinate human erythrocytes.
Strongest agglutination was obtained with the presence of intracellular extracts and rat erythrocytes of rabbits. The ISP2 environment better promotes the production of lectins. Intracellular fractions strongly caking rat erythrocytes were retained. That of Pru16
strain for purification and characterization and that of the Vic8 strain to study the
immunomodulatory effect.
Lectins, the intracellular fraction of strain Pru16 underwent a first primary purification
with ammonium sulphate and gel filtration G75. Differential precipitation showed that the
first three fractions exhibit HA activity. After filtration, two peaks were observed (peaks A
and B). Peak B shows no HA activity. However, the specific activity of the crude extract was
8.37 HU / mg, and it increased to 14.95 UH / mg after ammonium sulfate precipitation, and
20 UH / mg (picA) .after G75 gel filtration. The HA activity of this fraction remains at
alkaline pH and high concentrations of EDTA.
After column chromatography mucin sepharose 4B (second purification), two peaks
are observed. The proteic fraction 33 peak 2 has a high specific HA activity about twelve
times greater than that observed for the crude extract (8.37 111.49 against UH / mg), however
the first peak shows no HA activity. The affinity test conducted with the proteic fraction 33 of Peak 2 showed that the Pru16 strain binds specifically to protein. The SDS-PAGE analysis
showed that the hemagglutinin contained in the portion 33 of the P2 peak is located in the
band 16kDa The identification of proteins 16kDa band, after digestion and elution of peptides by LC, was top by tandem mass spectrometry proteomics comparing with the database related
strains.
The identified analysis of 6 different proteins similar, one of them has a high score,
with a molecular weight similar to that for protein band. This is the lysozyme C who did him
no HA activity. Our protein of 16kDa presents antimicrobial activity.
The enhancement of the phagocytic index of the corrected phagocytic index, and decreased half-life time in circulation of carbon particles in the blood indicate that the Vic8 strain of the crude extract possess one or more factors able to significantly boost the phagocytic activity of macrophages and reticuloendothelial system in mice.
