Biologie Résumé : In the first part of this study, two collections of wheat and one collection of triticale cultivated are analyzed to assess the genetic polymorphism of HMW-GS and SG-FPM: a collection of 69 triticale grown in France, another collection of 856 accessions of local durum wheat collected in Algeria and a priori divided into 17 botanical varieties according to their morphological characteristics, and the last two collections of 40 and 30 varieties of bread and durum wheats from Algerian oases. Biochemical analysis was performed by SDS-PAGE. In hexaploid triticale collection, allele nomenclature was compiled with a total of 36 alleles expressed at seven loci Glu-A1, Glu-B1, Glu-R1, Gli-R2, B2-Glu, Glu-A3, Glu- B3. This diversity has highlighted two major groups of triticale: winter and spring triticales.
Important variation was found in the collection of botanic durum wheat. Among the 16 alleles identified at the Glu-1 loci, two were new. The first named Glu-B1i1 encoding for two bands located between 17 and 18 which was assigned the nomenclature 171 and 181. The other named Glu-B1e1 codes two bands similar to 20x and 20y but with faster mobility, which were named 20x1-20y1. At Glu-3 and Glu-2 loci, 19 alleles were identified, where the allele named Glu-B3ab (encoding for subunits 2-8-9-13-16) was considered as new.
Analysis of Saharan wheats has identified a total of 32 and 29alleles for bread and durum wheat, respectively, which in combination resulted in 36 and 29 different patterns respectively. In bread wheat 3, 9 (including 1 new) and 4 alleles were observed at Glu-A1, Glu-B1 and Glu-D1 loci encoding HMW-GS respectively. LMW-GS displayed similar polymorphism, as 4, 9 and 3 alleles were identified at loci Glu-A3, Glu-B3 and Glu-D3 respectively. In durum wheat collection, 3 alleles were expressed at Glu-A1 locus and 9 (including 1 new) at Glu-B1. At Glu-A3, Glu-B3 and Glu-B2 loci 7, 8 (including 1 new) and 2 alleles were identified. Knowledge of these alleles is an important one hand to estimate the genetic variability and varietal identification and secondly in the selection for grain quality.
Another part of this study aimed to characterize the major allergens of wheat and / or cereals, to break down the specific IgE response to evaluate the role of different protein families in the allergenicity of wheat. The analysis included 10 varieties and 14 sera from allergic Algerian and French patients to wheat. All biochemical and immunological analysis showed that all proteins are recognized by patients regardless of clinical grade observed. 90% of the sera reacted fraction to albumins / globulins fraction in immunoblotting. IgE sera recognize more than one wheat protein and results can be observed that the immunoreactivity varies with genotypes. And the antigenic profile that emerges from these results is composed of five major antigens, albumins / globulins, /, and gliadins and glutenins subunits low molecular weight and a minor antigen subunits of glutenin high molecular weight. It is difficult to conclude on the type of sensitization to one or more common epitopes of these patients. Four protein bands were identified by proteomic analysis. Confronted by mass spectrometry analysis LC-MS/MS, we can suggest the existence of thirteen wheat allergens in the four groups analyzed.

Biologie Résumé : The purpose of our work has focused on the study of Teucrium polium L. Capitatum, which is a widespread medicinal plant in algerian steppes. Several study topics were investigated such as the morphology, physio-biochemical constituents and biological effects.
The morphological analysis consisted of ten populations sampled at different habitats in eastern Algeria. This study revealed that the leaves have the lowest number (five out of ten) of types of trichomes, followed by the calyx and corolla with eight types for each. This confirms the absence of intraspecific variability within the study populations. Furthermore, the analysis of relationships between morphological characteristics and environmental factors of their habitat showed that the latter influences the length of trichomes.
The use of one-dimensional electrophoresis technique on polyacrylamide gel in the presence of detergent (SDS-PAGE) allowed to determine the total protein profiles of the ten study populations. The similarity coefficients of polypeptide bands indicated high similarity between populations.
Biochemical analyzes of the three phenotypes of T. polium L. Capitatum showed that extracts of purple phenotypes were richest in anthocyanins, soluble sugars and carotenoids compared with pink and white phenotypes enlightening there by the importance of flower color.
The combination of certain chemical analysis methods, spectrophotometry (UV-visible) and chromatographic (HPLC) have identified three flavonoids including mostly the cyanidin and two other flavonoids, quercetin and caffeic acid.
The evaluation of the antioxidant potency, that was conducted using the method of trapping the free radical DPPH and that of iron reduction FRAP, revealed that the extract of Biskra population showed higher reducing capacity compared to that of Sétif, with a strong trapping of DPPH● radicals.
As regards to the antibacterial activity, it appears that extracts from two populations have acceptable antibacterial properties.
The pharmacological effect of T. polium L. Capitatum showed an interesting diuretic property.

This study is based on the variation of subunits glutenin high molecular weight HPM of 66 accessions of Aegilops belonging to the section Sitopsis, colled: Ae.bicornis, Ae.longissima, Ae.sharonensis, Ae.seassii and Ae.speltoides, using monodimensionnel electrophoresis. The presence of 3 glutenin bands in several accessions of Ae.longissima seems to be evidence for the existance of at least 3 genes encoding the SG-HPM in the genome S of Aegilops. The technic of SDS-PAGE has permitted us to estimate a high genetic variability of SG-HPM in Ae.speltoides. These results can be used in improving the technological quality of wheat. From the comparison between the glutenin patterns found in the species analyzed and those band observed for the B genome of hexaploid wheat, it can be concluded that Ae.bicornis and Ae.longissima are unlikely donors of the B genome.Note de contenu :

Biologie Résumé : In the first part of this study, several collections were analyzed in order to appreciate the allelic variation of high and low-molecular-weight (HMW and LMW) glutenin subunits composition: a collection of 59 accessions representing 5 genomes (U, C, M, D, N), collection of 856 accessions of local durum wheat (Triticum durum Desf.) collected in Algeria and divided a priori according to their agronomical and morphological traits in 17 varieties, a collection of thirty accessions of lines sisters coming from the ICARDA, obtained from interspecific crossings between 2 Syrian varieties of durum wheat (Cham and Oum Rabi) and 2 related species (T. dicoccum and T. polonicum) and two other collections of 71 bread wheat and 120 durum wheat germoplasm grown in Algeria. Seed samples were provided for the most part by the International Center for Agricultural Research in the Dry Areas (Alep, Syria). The biochemical analysis was carried out by monodimensionel SDS-PAGE.
In Aegilops collection, several new alleles corresponding to HMW subunits were detected and an allelic nomenclature was drawn up in which a total of 16 alleles were exprimed at the locus Glu-U1, 13 at Glu-M1, 9 at Glu-C1, 6 at Glu-N1 and 10 at the Glu-D1 locus.

In the collection of related durum wheat, we can count six allelic forms, which two are localised at the locus Glu-A1, it acts of the allele “a” and the allele “c” and four at the locus Glu-B1, they are “a, b, d and i”. While referring to international allelic nomenclature, these alleles code for following proteins: 1, nul, 7, 7+8, 6+8 and 17+18 respectively. Important polymorphism caracterise HMW-GS with a total of 13 alleles identified.The knowledge of these alleles constitutes an important tool for the identification varietal of wheat.
Extensive variation has been found in bread and durum wheat germoplasm grown in Algeria. In bread wheat 3, 6 and 5 alleles were observed at Glu-A1, Glu-B1 and Glu-D1 loci encoding HMW-GS respectively. LMW-GS displayed similar polymorphism, as 4, 9 and 3 alleles were identified at loci Glu-A3, Glu-B3 and Glu-D3 respectively. In durum wheat collection, 3 alleles were exprimeted at Glu-A1 locus and 9 at Glu-B1. At Glu-A3, Glu-B3 and Glu-B2 loci 5,8 and 2 alleles were identified.
The new HMW and LMW glutenin variation found in this work suggests their possible utilisation in breeding for wheat quality. The aim of the second part of this study was to evaluate the differences due to the culture regimes (biological versus conventional) in composition of total and metabolic proteins in the mature wheat grain. The analyses were carried out on three classes of sizes of grains (small, middle and large) from two varieties of bread wheat: Ataro and Renan. The two varieties exhibited differences on the grain sizes distribution. The one-dimensional electrophoretic analyses of storage and metabolic proteins showed a significant effect of the culture regime on HMW-SG and LMW-SG for the two varieties. Many gliadins (5 bands out of 16 for Renan and 11 out of 22 for Ataro) were influenced by culture regimes. The proteomic approach, carried out on the Ataro variety, revealed for all sizes of grains, 20 variable spots of which three were more expressed in biological regime. Analysis of the proteome between sizes revealed 149 spots quantitatively different from one class to another for the biological regime, against 115 for the conventional regime. Biologie Résumé : In the first part of this study, several collections were analyzed in order to appreciate the allelic variation of high and low-molecular-weight (HMW and LMW) glutenin subunits composition: a collection of 59 accessions representing 5 genomes (U, C, M, D, N), collection of 856 accessions of local durum wheat (Triticum durum Desf.) collected in Algeria and divided a priori according to their agronomical and morphological traits in 17 varieties, a collection of thirty accessions of lines sisters coming from the ICARDA, obtained from interspecific crossings between 2 Syrian varieties of durum wheat (Cham and Oum Rabi) and 2 related species (T. dicoccum and T. polonicum) and two other collections of 71 bread wheat and 120 durum wheat germoplasm grown in Algeria. Seed samples were provided for the most part by the International Center for Agricultural Research in the Dry Areas (Alep, Syria). The biochemical analysis was carried out by monodimensionel SDS-PAGE.
In Aegilops collection, several new alleles corresponding to HMW subunits were detected and an allelic nomenclature was drawn up in which a total of 16 alleles were exprimed at the locus Glu-U1, 13 at Glu-M1, 9 at Glu-C1, 6 at Glu-N1 and 10 at the Glu-D1 locus.
Important variation was found in the collection of botanic durum wheat. Among the 16 alleles identified at the Glu-1 loci, two were new. The first named Glu-B1i1 encoding for two bands located between 17 and 18 which was assigned the nomenclature 171 and 181. The other named Glu-B1e1 codes two bands similar to 20x and 20y but with faster mobility, which were named 20x1-20y1 and were analyzed to determine the number of thiol groups.
