The object of our work focused on the valorization of the Algerian flora. For this, firstly; we carried out a phytochemical study on Calligonum azel and Calligonum comosum which concerned mainly the biologically active compounds then we tested the antioxidant, antibacterial, antifungal and anti-inflammatory properties of all the organic extracts and some isolated compounds. Secondly; we studied the chemical composition and biological activities of Thymus ciliatus essential oil.
The study of aerial parts of C. azel and C. comosum, in order to know the active compounds responsible for their biological activity, leading to purification, chromatographic identification (CC, TLC and HPLC), and characterization using the different spectroscopic methods (1D and 2D NMR, MS and UV) of twenty one pure compounds (21) in which eighteen (18) compounds were identified.
The investigation of the biological activities of the extracts of Calligonum azel and Calligonum comosum showed that the best antioxidant activity was exhibited by the Chloroform and ethyl acetate extracts of C. azel and the Chloroform extract of C. comosum. The chloroform extracts of both species showed the best antibacterial activity. Chloroform extract from C. comosum has better antifungal activity against Candida albicans ATCC 2019 and Candida parapsilosis ATCC 22019.
From the isolated compounds, three have an antioxidant and anti-inflammatory activity. These compounds were: protocatachuic ethyl ester acid, -diamino (4-hydroxy benzene) butanoic acid and tamgernanetin.
The qualitative and quantitative analysis of essential oil of T. ciliatus allows the identification of 25 compounds representing 87.18 % of the total essential oil mainly. The GC and GC-MS analysis showed that this essential oils was mainly rich of thymol (54.26%), carvacrol methyl ether (1,373 %), o-cymene (5.72 %) carvacrol (1.373%)-cymen-7-ol (17.58 %), however it was the first time that -cymen-7-ol (17.58 %) was found as main component in the essential oils of T. ciliatus.
The evaluation of the antibacterial activity was assessed by disc diffusion method against bacterial strains. The essential oil exhibited a good antibacterial activity against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 43300 and Staphylocossus aureus.
All of our works confirms the medicinal properties of the Saharian species, and contributes to their scientific descriptions. This work concerned the phytochemical and biological study of two Algerian medicinal plants Thymus ciliatus (Desf) and Asteriscus graveolens (Forsk.) dc. belonging to the families Lamiaceae and Asteraceae ,respectively. This study led to the isolation of twenty seven products from which seventeen were identified including. Among the isolated compounds, 5 sesquiterpenes from which 3 were new, 6 flavonoids which 1 is new, 4 phenolic compounds and 2 monoterpenes. The structures of the isolated products were elucidated mainly by the combination of spectroscopic methods mainly: NMR techniques (1H, 13C, COSY, HSQC and HMBC), UV spectrophotometry, we also used high resolution mass spectrometry methods and comparison with literature data. The quantification of total phenols and total flavonoids contents of the EtOAc, nBuOH extracts of the aerial parts of Thymus ciliatus (Desf) were determined, EtOAc extract had the highest phenolic and flavonoid contents followed by n-butanol extract. This extracts showed an excellent antioxidant activity by DPPH test. The antioxidant activity of AcOEt and n-BuOH extracts of Asteriscus graveolens was estimated by the DPPH test and β-Carotene bleaching assay. The best result was obtained for the ethyl acetate extract, especially in the DPPH method, where its effect is greater than BHT and α-tocopherol used as standards. Both extracts and some isolated compounds of Asteriscus graveolens were tested for their cytotoxic effect in human cancer cell lines (HCT-116, DLD1 and HDFa). The results indicate that EtOAc extract and isolated compounds 9 α-hydroxyparthenolide, 9β-hydroxyparthenolide et 7,12-dihydroxy-6,7- dihydro-5,(6) E-dehydronerolidol showed significant activity compared to the n-BuOH extract and and the two new compounds. The chemical composition of the essential oil of aerial parts of Thymus ciliatus (Desf) Benth studied by GC-FID and GC-MS showed that this oil is mainly composed of elemol (6.80%), carvacrol (5.86%), γ-muurolene (5.18%), β-sesquiphellandrene (5.09%), bicyclogermacrene (5.04%), β-Pinene (4.49%) and curcumene (4.20%).Note de contenu : Annexes. In this thesis, we carried out a phytochemical study on three plants : Salvia buchananii Hedge, Cistanche phelypeae (L.) Coutinho and Anarrhinum pedatum Desf. 37 compounds were isolated by the use of different chromatographic methods of separation and purification. Amongthem, 17 were new molecules. The isolated compounds were classified into many groups : triterpenes, phenylethanoid, iridoids, flavonoids, phényléthanoids. GC and GC/MS analyses of the essential oil of S. buchananii showed that it was mainly represented by : 1,8-cineole (23.3%) α-pinene (19.8%), camphene (16.5%) and the β-pinene (11.5%). This oil showed a good antioxidant activity by the bleaching of β-carotene. The cytotoxic activity of the triterpenes SBR1 and SBR2 showed an interesting antiproliferative activity with similar potency in all cell lines HeLa. In addition, several compounds (CT1-CT7) showed a promising activity on hMAGL. This is the first report of naturally occurring phenyletanoid as a MAGL inhibitor. Moreover, the new iridoid compound (APD14) the A. pedatum exhibited the most potential antiangiogenic activity.
