The object of our work focused on the valorization of the Algerian flora. For this, firstly; we carried out a phytochemical study on Calligonum azel and Calligonum comosum which concerned mainly the biologically active compounds then we tested the antioxidant, antibacterial, antifungal and anti-inflammatory properties of all the organic extracts and some isolated compounds. Secondly; we studied the chemical composition and biological activities of Thymus ciliatus essential oil.
The study of aerial parts of C. azel and C. comosum, in order to know the active compounds responsible for their biological activity, leading to purification, chromatographic identification (CC, TLC and HPLC), and characterization using the different spectroscopic methods (1D and 2D NMR, MS and UV) of twenty one pure compounds (21) in which eighteen (18) compounds were identified.
The investigation of the biological activities of the extracts of Calligonum azel and Calligonum comosum showed that the best antioxidant activity was exhibited by the Chloroform and ethyl acetate extracts of C. azel and the Chloroform extract of C. comosum. The chloroform extracts of both species showed the best antibacterial activity. Chloroform extract from C. comosum has better antifungal activity against Candida albicans ATCC 2019 and Candida parapsilosis ATCC 22019.
From the isolated compounds, three have an antioxidant and anti-inflammatory activity. These compounds were: protocatachuic ethyl ester acid, -diamino (4-hydroxy benzene) butanoic acid and tamgernanetin.
The qualitative and quantitative analysis of essential oil of T. ciliatus allows the identification of 25 compounds representing 87.18 % of the total essential oil mainly. The GC and GC-MS analysis showed that this essential oils was mainly rich of thymol (54.26%), carvacrol methyl ether (1,373 %), o-cymene (5.72 %) carvacrol (1.373%)-cymen-7-ol (17.58 %), however it was the first time that -cymen-7-ol (17.58 %) was found as main component in the essential oils of T. ciliatus.
The evaluation of the antibacterial activity was assessed by disc diffusion method against bacterial strains. The essential oil exhibited a good antibacterial activity against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 43300 and Staphylocossus aureus.

The Extraction process has been done on the aerial parts of the two species previously mentioned using organic solvents with different methods followed by chromatographic separations (CC, TLC and HPLC), the outcomes of the said extraction gave 19 products of which 5 are new (2 saponins and 3 flavonol glycosides) which have been isolated from nbutanol phase of the hydroalcoholic extract of Gymnocarpos decander, and three known
compounds isolated from the ethyl acetate extract. The remaining 11 products isolated from Mentha pulegium are known flavonoids and phenolic acids.
Structural elucidation of the isolated compounds was performed by high resolution mass spectra (HRESI-MS), 1D NMR (1H, 13C) and 2D NMR (COSY, HSQC, HMBC) and comparing them with data from the literature.
The antiproliferative activity research tests of the five new products isolated from the n-butanol extract of G. decander showed that both saponins are active against Hela cell line and the results have been very promising and hence incite to effect the vivo studies to confirm its pharmacological properties.
Quantitative estimation for total polyphenols of the hydroalcoholic extract (aerial parts) of M. pulegium has shown clearly that this extract has a high content of polyphenols.
Evaluation of the biological activity carried out on this extract showed that this species had a good scavenging effect against the DPPH radical.
On the chemotaxonomic level, the results show a rich and varied composition of secondary metabolites which is matching the plants of the same family characteristics. In fact, for the Caryophyllaceae family, saponins are considered as chemotaxonomic markers. As for the Lamiaceae family, its reputation for flavonoid accumulation is well known.

Chromatographic separations and purifications of extracts from the aerial parts and roots of Mentha aquatica led to the isolation and identification of 1 6 compounds, including one new triterpene and 5 triterpene acids which are new for the genus.
The structures of isolated compounds were particularly established by the use of NMR 1D and 2D (1H, 13C, COSY H-H, HMQC and HMBC), high resolution mass spectrometry (ESI-MS and HR-ESI-MS) and by comparison with literature data.
GC and GC/MS analyses of T. dreatensis essential oil showed that it was mainly represented by thymol (28.1%) followed by γ-terpinene (18.7%), thymyl methyl ether (10.9%) and linalool (5.0%). This oil showed a good antioxidant activity by the bleaching of β-carotene.
5 methods were used to evaluate the antioxidant activity of Mentha aquatica and Stachys guyoniana. The latter showed a good activity with 4 methods: β-carotene, DPPH, ABTS and metal chelating whereas Mentha aquatica exhibited a moderate activity with CUPRAC and ABTS tests. Compared with the other tested extracts, the n-butanol extract of
Stachys guyoniana showed the highest anticholinesterase activity.
The two plants showed an antibacterial potential against some bacterial strains, by the use of agar diffusion method and MIC's determination.

Our work on the extraction of the aerial parts of G. numidica followed by the chromatographic separation permitted the isolation of 14 secondary metabolites from the chloroformic and ethyl acetate extracts. The structures of 13 were identified and fully described, including a new triterpene. The molecular structures of isolated compounds were established mainly by the use of NMR 1D and 2D (1H, 13C, TOCSY-1D, COSY, HSQC and HMBC), high resolution mass spectrometry (HRESI-MS) and by comparison with the literature Data. The antioxidant activity of CHCl3, EtOAc and n-BuOH extracts of G.numidica has been investigated by DPPH test, the results showed that the ethyl acetate extract has a good scavenging effect in vitro against the DPPH radical (IC50 = 40.43 ±3.09 µg / ml). The antiproliferative activity of isolates ( lignans and saponins) was investigated on Jurkat, HeLa and MCF-7 cell lines. The most active component, the triterpene, 3-O- β-D-glucopyranosyl-olean-12-en-3β,27,28,29-tetraol, showed activity in all cell lines, an IC50 value of 37 ± 2.3 µM (Jurkat), 35 ± 1.7 µM (HeLa) and 50 ± 3.6 µM (MCF-7) respectively. The mechanism underlying its antiproliferative effect was investigated in HeLa cells. The results showed that this compound affect the cell cycle progression by inducing a robust G2/M bloc in HeLa cells. Note de contenu : Annexe. The work presented in this thesis contributes to the valorization of both Algerian medicinal plants (Crataegus oxyacantha L. (Rosaceae) and Cytisus triflorus L'Her. (Fabaceae) by characterizing them by a phytochemical study, an evaluation of the antioxidant and antimicrobial activities, and a structural identification of certain phenolic compounds. Thus, in order to obtain an extract enriched in metabolites of interest, flavonoids, we opted for two extraction methods, a liquid solid extraction using different solvents of increasing polarity (CHCl3, EtOAc and n-Butanol) and a second extraction using the Matrix solid phase dispersion (MSPD) method, using different solvents of increasing polarity (100% H2O, 1:1 H2O / EtOH and 100% EtOH).
This study allowed the isolation 21 compounds and structural elucidation of 16 compounds.
Of these 16 compounds, 3 acids and 7 flavonoids: (3 flavonols including 1 glucosyl, 1 glucosyl flavanone, 1 Flavanol and 2 flavones) were isolated from C. oxyacantha L. From the EtOAc extract the aerial parts (Flowers + Leaves) of the species Cytisus triflorus L'Her., 6 secondary metabolites were identified (flavonoids). Among these compounds, 3 flavones: (Chrysin, Prunetin) and 3 glucosyl flavonoids (2 Chrysine and Isoquercitrin). Of these isolated compounds, 3 new in the species. These major active ingredients possess various biological activities and play an important and acknowledged role in maintaining good health. The structures of the isolated compounds were established by HPLC-DAD chromatogram analysis, UV-vis spectra and high resolution electrospray mass spectra (HRESI-MS), by extensive use and interpretation of NMR experiments spectra : 1D NMR (1H, 13C and DEPT) and 2D NMR (COSY, HSQC and HMBC) and by comparison with the literature data. The in vitro evaluation antioxidant activity of the various extracts and of three isolated products of C. oxyacantha, and the AcOEt extract and three products isolated from C. triflorus was performed by the radical scavenging method (DPPH *). The extracts of two plants have interesting antioxidant activities, dependent on the content of total polyphenols and flavonoids. The various extracts studied were also revealed a high antibacterial activity towards S. aureus. Similarly, a tendency shown indicating that the richest extracts of flavonoids and polyphenols exhibit the higher antibacterial activities.Note de contenu :
