The works presented in this manuscript is a continuation of the phytochemical research carried out by our phytochemistry group affiliated to the VARENBIOMOL research unit from many years on medicinal and aromatic plants growing in different regions of the country, including the vaste Sahara. The first aim of this research is to value the biological resources related to the plant biodiversity of the Algerian flora and the knowledge associated with this flora in terms of traditional use in medicine, and the second one is the separation and identification of new bioactive compounds with interesting biological activities. The choice of Asphodelus tenuifolius specie for a phytochemical and a biological study in this thesis was based mainly on the fact that this specie belongs to the Liliaceae family known for its richness in various secondary metabolites with a good biological and pharmacogical interest, as well as for the good reputation of using this plant in local traditional medicine. For this purpose, the organic extracts of A. tenuifolius specie were studied chemically using various chromatographic methods (TLC, CC, preparative plates, HPLC…) to lead the isolation and the identification of seventeen pure compounds using various spectroscopic analysis, 1D and 2D NMR (1H, 13C, COSY, HSQC, HMBC ...), ECD analysis, HRESI-MS and ESI-MS spectroscopy, UV-Visible, IR and also with comparison with the literature data. All These data allowed us especially to characterize two new structures of anthraquinones isolated from the Chloroform extract with ten other known compounds dividing into two flavonoids, one sterol, one nitrogen phenolic compound, an anthraquinone and two glycoside anthraquinones with three more compounds in the process of identification, most of these classes are known with their therapeutic benefit and interesting biological activities. Phytochemical study of Ethyl Acetate extract of this specie allowed also the isolation of six known compounds: two phenolic acids, three flavonoids and one anthraquinone. Quantitative estimation of total polyphenols and flavonoids contents (TPC and TFC) showed that the Chloroform extract has a high content of polyphenols and flavonoids compared to Ethyl acetate and Butanol extracts. The biological activities assays like: antioxidant, antibacterial, antifungal, cytotoxic and antiviral performed for the different organic extracts, as well as a cytotoxic activity in A375 (human melanoma) cancer cell line carried out on the two new isolated compounds, confirm the pharmacological and the therapeutic properties of this specie and give an explication of their traditional uses.

and biological studies on the plants of the genus Centaurea (Asteraceae). The species
of this genus are potential reserves of bioactive secondary metabolites.
The present work concerned initially the in vitro evaluation of the cytotoxic,
antifungal and direct and indirect antimicrobial activities of the chloroform soluble part of
the hydro-alcoholic extract of the aerial parts of Centaurea diluta Ait. subsp. algeriensis
(Coss. & Dur.) Maire, an endemic species to Algeria and Morocco. According to an exhaustive
bibliographical research carried out on Centaurea species, no phytochemical nor biological report
is available on this subspecies so far; which constitutes one of our selection criteria for this plant.
Inactive against 7 tested microorganisms (plant pathogens) fungi, bacteria and yeast, and
also inactive in the direct antibacterial test, this extract displayed significant cytotoxic effects on
cancer cells (the A549 non-small-cell lung carcinoma, the MCF7 breast adenocarcinoma and the
U373 glioblastoma). In indirect antibacterial tests, the CHCl3 extract was able to potentiate the
effect of -lactam antibiotics on methicillin-resistant Staphylococcus aureus (MRSA), reducing
the minimal inhibitory concentrations (MICs) by a factor of 2-32-fold. No synergy was found
between the extract and streptomycin. Biological data allowed to guide the chemical composition
of the extract by fractionation, separation and purification on silica gel 60 (CC and TLC).
This study led to the isolation of nine components in a pure state and the identification of
five of them, by the combination of several spectroscopic analyses. These products were
two phenolic compounds (vanillin 1, paridol 3), a lignane [(-arctigenin 2] and two flavonoid
aglycones (eupatilin 4 and jaceosidin 5). All the isolated compounds are described for the first
time for this species. The 5 identified compounds were evaluated for cytotoxic and antimicrobial
activities. Moderate cytotoxic effects were observed on the six tested cancer cell lines (A549,
MCF7, U373, Hs683 human glioma, PC3 human prostate and B16-F10 murine melanoma)
for both the flavonoid aglycones and the lignane with IC50s in the range 28 μM to 40 μM.
In the direct antibacterial test by TLCbioautography, among the five isolated
compounds, only jaseosidin 5 showed a moderate antimicrobial activity but this
activity was relatively weak on the positive and negative Gram bacteria (MIC of 200 μg/mL
on MRSA C98506, MRSA C100459, MRSA ATCC33591, MSSA ATCC6538, E. coli
ATCC25922).

The present work aims to the valorization of Algerian flora by phytochemical et biological investigations on two species : Iris unguicularis Poir. and Dittrichia viscosa (L.) Greuter. This study allowed to identify several phytochemical compounds and to record satisfactory results for different biological activities tested for this two species. LC-MS IT-TOF analysis of the different extracts (AI, CI et BI) of Iris unguicularis Poir. species led to the quantification of 70 compounds, while the LC-MS/MS analysis accomplished by a previously validated method by using 37 standards revealed the presence of 17 phenolic compounds and the richest extracts were, AI and CI, AI extract was mainly composed by: 4-OH-benzoic acid (2218.2 µg analyte/g extract), syringinic acid (1218.6 µg analyte/g extract), protocatechuique acid (1080. 4 µg analyte/g extract) and p-coumaric acid (1071.2 µg analyte/g extract), the CI extract was mainly composed by: naringinia (1453.4 µg analyte/g extract) and ferulic acid (1088, 41 µg analyte/g extract). Results showed also the high antioxidant an antibacterial activities of these extracts . Structural elucidation of isocostic acid isolated from the species Dittrichia viscosa (L.) Greuter, was performed by NMR technique (1H and 13C). The results of activities carried out on this compound revealed its apoptotic effect by acting as pro-oxidant which allows to generate the superoxide anion in one hand and on the other hand by its effect on mitochondrial swelling. The viability of cells via MTT assay tested against U118, Caco-2, SKOV-3 and HDF lines showed that isocostic acid had a significant effect on cell proliferation by causing a dose-dependent cytotoxicity especially against U118 line in which proliferation is clearly decreased between 100μg and 200 μg with an IC50 of 72.32 μg/ml.

These compounds were identified by the combination of spectroscopic methods including: UV-visible absorption spectroscopy, high resolution mass spectrometry, RMN 1D and 2D, as well as comparison with literature data.
The pure compounds and the ethyl acetate fraction of F. longipes Coss. showed an excellent antioxidant activity compared to the standards. The petroleum and butanolic fractions of the species Athamanta sicula L. showed a moderate anticholinesterase activity against AChE. The pure compounds of the species Athamanta sicula L. showed a moderate cytotoxic activity.

Our work on the extraction of the aerial parts of G. numidica followed by the chromatographic separation permitted the isolation of 14 secondary metabolites from the chloroformic and ethyl acetate extracts. The structures of 13 were identified and fully described, including a new triterpene. The molecular structures of isolated compounds were established mainly by the use of NMR 1D and 2D (1H, 13C, TOCSY-1D, COSY, HSQC and HMBC), high resolution mass spectrometry (HRESI-MS) and by comparison with the literature Data. The antioxidant activity of CHCl3, EtOAc and n-BuOH extracts of G.numidica has been investigated by DPPH test, the results showed that the ethyl acetate extract has a good scavenging effect in vitro against the DPPH radical (IC50 = 40.43 ±3.09 µg / ml). The antiproliferative activity of isolates ( lignans and saponins) was investigated on Jurkat, HeLa and MCF-7 cell lines. The most active component, the triterpene, 3-O- β-D-glucopyranosyl-olean-12-en-3β,27,28,29-tetraol, showed activity in all cell lines, an IC50 value of 37 ± 2.3 µM (Jurkat), 35 ± 1.7 µM (HeLa) and 50 ± 3.6 µM (MCF-7) respectively. The mechanism underlying its antiproliferative effect was investigated in HeLa cells. The results showed that this compound affect the cell cycle progression by inducing a robust G2/M bloc in HeLa cells. Note de contenu : Annexe.
