This work is devoted to phytochemical and biological studies of two Algerian species, Pituranthos battandieri Maire (Apiaceae), an endemic plant of the Algerian-Moroccan Sahara and Frankenia thymifolia Desf. (Frankeniaceae) endemic of North Africa. The endemism of these species has encouraged us to conduct this work. Our objective consists of the isolation and structural determination of bioactive molecules starting from various extracts of these two plants as well as the in vitro evaluation of their antiparasitic, antibacterial and antioxidant activities. The cytotoxicity on murine macrophage of J 774 cell line of the antiparasitic compounds isolated from P. battandieri by bio-guided fractionation, separation and purification was also evaluated. The evaluation of the antiparasitic activity of the extracts from aerial parts and roots (taken separately) of P. battandieri against Leishmania amazonensis and donovani, Trypanosoma cruzi and Acanthamoeba castellanii showed significant antiparasitic properties. The bio-guided fractionation of the most active extracts (dichloromethane and petroleum ether from aerial parts and the roots, respectively) by various chromatographic techniques, led to the isolation of 14 active compounds on L. amazonensis and T. cruzi, of which 7 (P1-P7) have been identified. These compounds were 2 polyynes including 1 new as a natural product, 3 furanocoumarins, 1 monoterpene and 1 phenolic compound. Among the tested compounds, (-)-falcarindiol (P7) and (-)-panaxydiol (P6) exhibited significant antileishmanial and antitrypanosomal activities with low toxicity to J 774 macrophages. The antioxidant activities of the various extracts of this species were evaluated by several methods. The dichloromethane and ethyl acetate extracts of the aerial parts and petroleum ether of the roots showed remarkable antioxidant effects compared to the standards used. The phytochemical investigation of AcOEt and n-BuOH extracts from the aerial parts of F. thymifolia led to the isolation of 12 compounds, of which 7 flavonoids and a gallic acid derivative
were characterized. The structural elucidation of the compounds was carried out by the combination of their spectroscopic data: (UV-Vis), 1D-NMR (1H, 13C and DEPT), 2D-NMR (COSY, HSQC, HMBC, and GOESY), high resolution mass spectrometry with EI and ESI ionization modes (HR-EI-MS and HRESIMS) and by comparison with literature data.
The antioxidant powers of CHCl3, AcOEt and n-BuOH extracts from F. thymifolia as well as those of the isolated compounds F1, F2 and F6 were evaluated by several methods: radical DPPH, radical-cation ABTS, bleaching of β-carotene, radical- anion superoxide, ferrous chelating power, CUPRAC and FRAP. The results clearly indicated high antioxidant potentials of the extracts and compounds tested. These results were in good agreement with the total polyphenol and flavonoid contents data. Moreover, the results of the evaluation of the antiparasitic activity showed that the AcOEt extract
showed a remarkable antiparasitic effect compared to n-BuOH and CHCl3 extracts, against A. castellanii and L. amazonensis parasites. While, AcOEt and n-BuOH extracts showed moderate antibacterial activity on the bacterial strains tested.Note de contenu :

The works presented in this manuscript is a continuation of the phytochemical research carried out by our phytochemistry group affiliated to the VARENBIOMOL research unit from many years on medicinal and aromatic plants growing in different regions of the country, including the vaste Sahara. The first aim of this research is to value the biological resources related to the plant biodiversity of the Algerian flora and the knowledge associated with this flora in terms of traditional use in medicine, and the second one is the separation and identification of new bioactive compounds with interesting biological activities. The choice of Asphodelus tenuifolius specie for a phytochemical and a biological study in this thesis was based mainly on the fact that this specie belongs to the Liliaceae family known for its richness in various secondary metabolites with a good biological and pharmacogical interest, as well as for the good reputation of using this plant in local traditional medicine. For this purpose, the organic extracts of A. tenuifolius specie were studied chemically using various chromatographic methods (TLC, CC, preparative plates, HPLC…) to lead the isolation and the identification of seventeen pure compounds using various spectroscopic analysis, 1D and 2D NMR (1H, 13C, COSY, HSQC, HMBC ...), ECD analysis, HRESI-MS and ESI-MS spectroscopy, UV-Visible, IR and also with comparison with the literature data. All These data allowed us especially to characterize two new structures of anthraquinones isolated from the Chloroform extract with ten other known compounds dividing into two flavonoids, one sterol, one nitrogen phenolic compound, an anthraquinone and two glycoside anthraquinones with three more compounds in the process of identification, most of these classes are known with their therapeutic benefit and interesting biological activities. Phytochemical study of Ethyl Acetate extract of this specie allowed also the isolation of six known compounds: two phenolic acids, three flavonoids and one anthraquinone. Quantitative estimation of total polyphenols and flavonoids contents (TPC and TFC) showed that the Chloroform extract has a high content of polyphenols and flavonoids compared to Ethyl acetate and Butanol extracts. The biological activities assays like: antioxidant, antibacterial, antifungal, cytotoxic and antiviral performed for the different organic extracts, as well as a cytotoxic activity in A375 (human melanoma) cancer cell line carried out on the two new isolated compounds, confirm the pharmacological and the therapeutic properties of this specie and give an explication of their traditional uses.

تقنية الرنين النووي المغناطيسي تربينات نشاطات بيولوجية زيوت طيارة"Index. décimale : 540 Chimie et sciences connexes Résumé : This present work concerns the phytochemical and biological study of three Algerian plants belonging to the Lamiaceae family : Mentha aquatica L., Stachys guyoniana de Noé and Thymus dreatensis Batt. Chromatographic separations and purifications of extracts from the aerial parts and roots of Mentha aquatica led to the isolation and identification of 1 6 compounds, including one new triterpene and 5 triterpene acids which are new for the genus.
The structures of isolated compounds were particularly established by the use of NMR 1D and 2D (1H, 13C, COSY H-H, HMQC and HMBC), high resolution mass spectrometry (ESI-MS and HR-ESI-MS) and by comparison with literature data.
GC and GC/MS analyses of T. dreatensis essential oil showed that it was mainly represented by thymol (28.1%) followed by γ-terpinene (18.7%), thymyl methyl ether (10.9%) and linalool (5.0%). This oil showed a good antioxidant activity by the bleaching of β-carotene.
5 methods were used to evaluate the antioxidant activity of Mentha aquatica and Stachys guyoniana. The latter showed a good activity with 4 methods: β-carotene, DPPH, ABTS and metal chelating whereas Mentha aquatica exhibited a moderate activity with CUPRAC and ABTS tests. Compared with the other tested extracts, the n-butanol extract of
Stachys guyoniana showed the highest anticholinesterase activity.
The two plants showed an antibacterial potential against some bacterial strains, by the use of agar diffusion method and MIC's determination.

ABTS + test, followed by fractions F40:60 and F80:20. Furthermore, the F60:40 fraction of
the hydromethanolic extract of the cones of A. numidica was the best inhibitor of the enzyme
acetylcholinesterase whereas the F80:20 fraction showed the highest activity against
butyrylcholinesterase. The extracts of the two studied plants and the isolated compounds
showed a good antimicrobial activity against a large spectrum of Gram-positive bacteria and
on the tested fungi (Candida)

ABTS + test, followed by fractions F40:60 and F80:20. Furthermore, the F60:40 fraction of
the hydromethanolic extract of the cones of A. numidica was the best inhibitor of the enzyme
acetylcholinesterase whereas the F80:20 fraction showed the highest activity against
butyrylcholinesterase. The extracts of the two studied plants and the isolated compounds
showed a good antimicrobial activity against a large spectrum of Gram-positive bacteria and
on the tested fungi (Candida)
