and biological studies on the plants of the genus Centaurea (Asteraceae). The species
of this genus are potential reserves of bioactive secondary metabolites.
The present work concerned initially the in vitro evaluation of the cytotoxic,
antifungal and direct and indirect antimicrobial activities of the chloroform soluble part of
the hydro-alcoholic extract of the aerial parts of Centaurea diluta Ait. subsp. algeriensis
(Coss. & Dur.) Maire, an endemic species to Algeria and Morocco. According to an exhaustive
bibliographical research carried out on Centaurea species, no phytochemical nor biological report
is available on this subspecies so far; which constitutes one of our selection criteria for this plant.
Inactive against 7 tested microorganisms (plant pathogens) fungi, bacteria and yeast, and
also inactive in the direct antibacterial test, this extract displayed significant cytotoxic effects on
cancer cells (the A549 non-small-cell lung carcinoma, the MCF7 breast adenocarcinoma and the
U373 glioblastoma). In indirect antibacterial tests, the CHCl3 extract was able to potentiate the
effect of -lactam antibiotics on methicillin-resistant Staphylococcus aureus (MRSA), reducing
the minimal inhibitory concentrations (MICs) by a factor of 2-32-fold. No synergy was found
between the extract and streptomycin. Biological data allowed to guide the chemical composition
of the extract by fractionation, separation and purification on silica gel 60 (CC and TLC).
This study led to the isolation of nine components in a pure state and the identification of
five of them, by the combination of several spectroscopic analyses. These products were
two phenolic compounds (vanillin 1, paridol 3), a lignane [(-arctigenin 2] and two flavonoid
aglycones (eupatilin 4 and jaceosidin 5). All the isolated compounds are described for the first
time for this species. The 5 identified compounds were evaluated for cytotoxic and antimicrobial
activities. Moderate cytotoxic effects were observed on the six tested cancer cell lines (A549,
MCF7, U373, Hs683 human glioma, PC3 human prostate and B16-F10 murine melanoma)
for both the flavonoid aglycones and the lignane with IC50s in the range 28 μM to 40 μM.
In the direct antibacterial test by TLCbioautography, among the five isolated
compounds, only jaseosidin 5 showed a moderate antimicrobial activity but this
activity was relatively weak on the positive and negative Gram bacteria (MIC of 200 μg/mL
on MRSA C98506, MRSA C100459, MRSA ATCC33591, MSSA ATCC6538, E. coli
ATCC25922).

(Zygophyllaceae) and the chemical composition and antioxidant activity study of fruit essential oil of Zizyphus lotus (L.) Desf. (Rhamnaceae).
In a first part, the phytochemical investigation of the species F. longispina using different chromatographic methods (CC, TLC and HPLC) led to isolation and identification by different spectroscopic methods (1D and 2D, MS and UV) 18 compounds. Among these isolated compounds, a new monoterpene ∆-2-carene-6-ol-6-O-(2',6'-diacetyl)-β-D-glucopyranoside, together with four other monoterpenes, seven flavonoids, four phenolic derivatives and two
sterol.
Moreover, the in vitro free radical-scavenging activity of the all extracts has been investigated by DPPH test, which have demonstrated that ethyl acetate extract are good sources of antioxidant compounds. However, all extracts of F. longispina showed no substantial result compared to the antioxidant activity of ascorbic acid. In addition, the total phenolic and the flavonoids contents were measured using spectrophotometers methods, the results showed that the acetate extract has a high content compared to the other extracts. As for antibacterial activity study, showed that a high antibacterial effect of the ethyl acetate extract.
In a second part, the chemical composition of fruit essential oil Z. lotus has been investigated by the GC-FID and GC-MS analysis, which led to identified 38 compounds. Fatty acids represented the major fraction (saturated and unsaturated compounds 67.8 and 11%, respectively), followed by hydrocarbons (10.8%) and sesquiterpenes (1.1%).These results were quite different from those obtained previously on the fixed oil of the fruit of this species from two samples from Algeria.
The antioxidant property of this oil was evaluated using β-carotene bleaching method. The result indicated the potent antioxidant activity of essential oils from Z. lotus. The purpose of this work is the phytochemical and pharmacological investigation that led to the isolation of the molecules responsible for the radical scavenging capacity of this species. The CHCl3, AcOEt and n-BuOH soluble parts of the hydro-ethanol extract (8:2) of the aerial parts were evaluated for their free radical scavenging capacity by on-line (HPLC-ABTS•+) assay. The obtained data were confirmed off line by TEAC and ORAC assays. A subsequent fractionations by HPLC reverse phase followed by spectroscopic analysis (HRMS, UV, NMR: 1H, 13C, COSY, HSQC and HMBC) were applied to identify the structure of the active compounds. Twenty three compounds of different categories were identified, among which nine phenolic acids: ferulic acid ethyl ester 2, p-coumaric acid ethyl ester 6, caffeic acid ethyl ester 10, vanillic acid 11, ferulic acid 12, alfrutamide 13, 4-hydroxy -benzoic acid 14, p-coumaric acid 15,caffeic acid 16, two phytosterols: stigmasterol 3 and beta-sitosterol 4 as a mixture (73% and 27%, respectively), four coumarins: 7-O-prenylscopoletin 5, isoscopoletin 7, scoparone 8 and scopoletin 9, two quinic acids: 3,5-di-caffeoylquinic acid 21 and 4,5-di-caffeoylquinic acid 22 and six flavonoids: chrysoeriol 1, astragalin 17, vicenin-2 18, isoscutellarein 6-C--Dglucopyranosyl 19, nicotiflorin 20 and vitexin 23. All the extracts showed a significant antioxidant activity on-line. The most active compounds were caffeic acid ethyl ester 10, caffeic acid 16, 3,5-di-caffeoylquinic acid 21 and 4,5- icaffeoylquinic acid 22. These results were validated off-line by ORAC and TEAC assays. Four compounds: 1, 5, 18 and 19 were described for the first time from the Convolvulaceae family, whereas compounds 2, 6, 8, 10, 13 and 21 were new for the genus Convolvulus. These results show a rich chemical composition and varied in secondary metabolites more precisely the phenolic compounds which have exhibited a very high antioxidant activity.

This work deals with phytochemical and pharmacological study of two species Salvia phlomoides Asso. from Lamiaceae family and Ferula vesceritensis Coss. Et Dur. from the Apiaceae family. The separation and purification by chromatographical methods of chloroform and ethyl acetate extract of Salvia phlomoides led to the isolation of nine compounds including four triterpenoids from which three are new, four flavonoids, one phenolic acid, and three flavonoids from the ethyl acetate extract of Ferula vesceritensis. The structural etermination of the isolated compounds was established on the basis of spectral analysis IR; ESI-HR-MS, 1D and 2D NMR (1H NMR, 13C NMR, DEPT-135, HSQC, HMBC, COSY and NOESY), and comparison with literature data. Evaluation of the cytotoxicity of the new triterpenoids showed a selective inhibitory effect against the panel of human tumor cell lines NCI-60 of the National Cancer Institute of the United States of America. Evaluation of the antioxidant activity by different methods of the ethyl acetate and n-butanol extracts of Salvia phlomoides and ethyl acetate extract of Ferula vesceritensis indicates an important antioxidant capacity. The GC and GC-MS analysis of the essential oils of the two species showed that the essential oil of Salvia phlomoides (studied for the first time) was mainly represented by Hexahydrofarnesyl acetone, Dibutyle phtalate, Phytol, α-Humulene, Caryophyllene Oxyde and β-Caryophyllene, and major components of the essential oil of Ferula vesceritensis (South) were Shyobunol, δ-Cadinene, ϒ-Cadinene, Isoledene and Dauca-5,8-diene, while for Ferula vesceritensis (East), D-Carvone and Rotundifolone were found as major components for the first time in the genus Ferula.Note de contenu :
