The Extraction process has been done on the aerial parts of the two species previously mentioned using organic solvents with different methods followed by chromatographic separations (CC, TLC and HPLC), the outcomes of the said extraction gave 19 products of which 5 are new (2 saponins and 3 flavonol glycosides) which have been isolated from nbutanol phase of the hydroalcoholic extract of Gymnocarpos decander, and three known
compounds isolated from the ethyl acetate extract. The remaining 11 products isolated from Mentha pulegium are known flavonoids and phenolic acids.
Structural elucidation of the isolated compounds was performed by high resolution mass spectra (HRESI-MS), 1D NMR (1H, 13C) and 2D NMR (COSY, HSQC, HMBC) and comparing them with data from the literature.
The antiproliferative activity research tests of the five new products isolated from the n-butanol extract of G. decander showed that both saponins are active against Hela cell line and the results have been very promising and hence incite to effect the vivo studies to confirm its pharmacological properties.
Quantitative estimation for total polyphenols of the hydroalcoholic extract (aerial parts) of M. pulegium has shown clearly that this extract has a high content of polyphenols.
Evaluation of the biological activity carried out on this extract showed that this species had a good scavenging effect against the DPPH radical.
On the chemotaxonomic level, the results show a rich and varied composition of secondary metabolites which is matching the plants of the same family characteristics. In fact, for the Caryophyllaceae family, saponins are considered as chemotaxonomic markers. As for the Lamiaceae family, its reputation for flavonoid accumulation is well known.

Our phytochemical and biological work concerns the aerial parts of two endemic Saharan species belonging to the Fabaceae family: Ononis angustissima Lam. Var and Lotus corniculatus L. Several chromatographic methods were used in our experimental work which allowed us to isolate nineteen products from the butanolic extract of Ononis angustissima Lam. Var. from which two are new, and eight products from Lotus corniculatus L. The structures were established by different spectral methods: nuclear magnetic resonance with different applications (NMR 1H, NMR 13C, DEPT, COSY, HMQC and HMBC), the mass spectrometry, UV, as well as comparison with literature data. The antiproliferative activity tests of the n-butanol extract and the compounds isolated from L. corniculatus were carried out on three cell lines: J774.A1 (murine macrophages monocytes), WEHI-164 (murine fibrosarcoma) and HEK-293 (human epithelial renal cells) have shown that the n-butanol extract and the pharbitoside A compound are the most active against the three cell lines. The two extracts and most of the isolated compounds of Ononis angustissima Lam. Var have excellent activity, so these molecules are considered first class antioxidants compared to reference molecules. They can therefore be used for therapeutic applications. Finally, the quantitative estimate of total polyphenols has shown that these extracts have a high content of polyphenols.Note de contenu :

The works presented in this manuscript is a continuation of the phytochemical research carried out by our phytochemistry group affiliated to the VARENBIOMOL research unit from many years on medicinal and aromatic plants growing in different regions of the country, including the vaste Sahara. The first aim of this research is to value the biological resources related to the plant biodiversity of the Algerian flora and the knowledge associated with this flora in terms of traditional use in medicine, and the second one is the separation and identification of new bioactive compounds with interesting biological activities. The choice of Asphodelus tenuifolius specie for a phytochemical and a biological study in this thesis was based mainly on the fact that this specie belongs to the Liliaceae family known for its richness in various secondary metabolites with a good biological and pharmacogical interest, as well as for the good reputation of using this plant in local traditional medicine. For this purpose, the organic extracts of A. tenuifolius specie were studied chemically using various chromatographic methods (TLC, CC, preparative plates, HPLC…) to lead the isolation and the identification of seventeen pure compounds using various spectroscopic analysis, 1D and 2D NMR (1H, 13C, COSY, HSQC, HMBC ...), ECD analysis, HRESI-MS and ESI-MS spectroscopy, UV-Visible, IR and also with comparison with the literature data. All These data allowed us especially to characterize two new structures of anthraquinones isolated from the Chloroform extract with ten other known compounds dividing into two flavonoids, one sterol, one nitrogen phenolic compound, an anthraquinone and two glycoside anthraquinones with three more compounds in the process of identification, most of these classes are known with their therapeutic benefit and interesting biological activities. Phytochemical study of Ethyl Acetate extract of this specie allowed also the isolation of six known compounds: two phenolic acids, three flavonoids and one anthraquinone. Quantitative estimation of total polyphenols and flavonoids contents (TPC and TFC) showed that the Chloroform extract has a high content of polyphenols and flavonoids compared to Ethyl acetate and Butanol extracts. The biological activities assays like: antioxidant, antibacterial, antifungal, cytotoxic and antiviral performed for the different organic extracts, as well as a cytotoxic activity in A375 (human melanoma) cancer cell line carried out on the two new isolated compounds, confirm the pharmacological and the therapeutic properties of this specie and give an explication of their traditional uses.

The chloroform extract was submitted to separation and purification by column and thin layer chromatography on silica gel (CCM).
The isolation and structure determination of isolated compounds were based on the different chromatographic methods(UV-Vis spectroscopy data, mass spectrometry,ID RMN (1H,13C)and 2D RMN experiments(COSY,HSQC,HMBC,HMQC and NOESY)
.
This work led to the isolation in the native state of 5 compounds which were :
2',4'-Dihydroxychalcone.
2'5,',6'Trihydroxychalcone.
6-Hydroxy -aurone.
5-Hydroxy,7Metxoy-flavone.
7Hydroxy,5Metoxy-flavone.
