Biologie Résumé : The work presented in this thesis contributes to the valorization of the aerial part of an endemic plant collected from the Algerian Sahara, Heliotropium undulatum Vahl (the family Boraginaceae). This study was conducted to evaluate the antioxidant and antiinflammatory activities of n-BuOH extracts of stems and leaves of H. undulatum using a rat- hydrazine model. In vitro studies (DPPH°, Chelation, FRAP, °OH) reveal that the n-BuOH extract of leaves has an antioxidant property. In vivo studies have also shown that the n-BuOH extract of 200 mg/kg leaves has demonstrated hepatoprotective activity by preserving liver function, modulating CYP2E1, neutralizing hepatic oxidative stress, and providing intra- mitochondrial redox status exhibited during treatment with hydrazine (300 mg/kg). The antiinflammatory activity was better pronounced by the n-BuOH extract of the stems, the demonstration of which is carried out using various in vitro tests (denaturation of albumin, HRBC, NO° scavenger). The study of modulatory pathways (hepatic carbonyl proteins, MPO, levels of NO, IL-ip and TNF-a) confirmed the in vivo anti-inflammatory potential of the n- BuOH extract of stems. The set of concomitant antioxidant and anti-inflammatory mechanisms characterizing the two extracts of Heliotropiuin undulatum is correlated with the qualitative and quantitative nature of the phenolic acids and flavonoids revealed via HPLC- MASS analysis and with the synergetic effect of all the phytoconstituents contained in this plant

(Boraginaceae) and Lifago dielsii Schwein. & Musch. (Asteraceae).
This phytochemical study led the isolation and identification of twenty products, seventeen of which were monoterpenes megastigans and phenolic type, were obtained from the chloroform and methanol extracts of the aerial parts of Heliotropium bacciferum Forssk.
While three glycosylated flavonoids were obtained from the ethyl acetate extract of the aerial parts of Lifago dielsii Schwein & Musch. The structures of the isolated products were elucidated mainly by the use of NMR techniques (1H, 13C, COSY, HSQC and HMBC), UV spectrophotometry and by comparison with those reported in the literature.
The quantification of the total phenolic content of the MeOH extract of H. bacciferum was carried out using the Folin-Ciocalteu method. This step showed a polyphenolic content of 68.0 mgGAE/g extract. This extract was evaluated for its antioxidant activity towards the free radical DPPH and the radical ABTS. The results showed that this extract exhibited a significant activity and is dependent on the concentration in vitro with respect to the DPPH
radicals (IC50 70.912 μg/ml), and ABTS (IC50 1.466 μg/ml), related to the presence of the phenolic derivatives.
Both extracts and some isolated products of H. bacciferum were tested for their cytotoxic effect in human cancer cell lines (HCT-116, DLD1 and HDFa). Only the chloroform extract showed a significant and concentration-dependent inhibitory effect on the growth of IC50 treated cancer cell lines of 0.095 mg/ml on HCT116 and 0.062 mg/ml on DLD1.
The quantification of total phenols and total flavonoids contents of the CHCl3, EtOAc, nBuOH extracts and of the methanol insoluble part (MeOH) of the aerial parts of L. dielsii was carried out using colometric methods. This study showed a phenolic content more important in the EtOAc extract. However, total flavonoids content was higher in n-BuOH extract (88.81%). The antioxidant activity of L. dielsii extracts was evaluated by the DPPH free radical scavenging and the lipid peroxidation inhibition (LPO) assays. The EtOAc extract has the strongest effect on DPPH with (IC50 = 47.80 ± 2.20 μg/ml) and the inhibition of lipid peroxidation (IC50 = 113.24 ± 0.65 μg/ml).

They are Centaurea choulettiana Pomel and Centaurea fragilis Dur.
The phytochemical investigation of cyclohexane, ethyl acetate and n-butanol extracts from the aerial parts of C. choulettiana and n-butanol extract of C. fragilis allowed the isolation and identification of 13 products. The structural elucidation of the isolated compounds was carried out by combining the various spectroscopic methods (UV, 1D & 2D NMR, HPLC/TOF-MS and GC/MS) as well as by comparing the results with the data of the literature.
The analysis of essential oil (flowers) of C. choulettiana by the GC/MS technique, carried out for the first time, showed the presence of several chemical groups from which, phthalate (18.38%) followed by β-eudesmol (6.8%), 5-epoxysalvial-4(14)-ene (6.6%), caryophyllene oxide (4.28%), tricosane (3.31%), 7-epi-α-eudesmol (3.08%) and heptacosane. The antibacterial activity of this oil was determined against five bacterial strains according to the disk diffusion method, the results showed modest activity against all the strains.
Quantification of the total polyphenols and total flavonoids of the EtOAc and n-BuOH (leaves and flowers) extracts of C. choulettiana was performed by colorimetric methods. This step showed a content of polyphenols and flavonoids more important in the AcOEt extract of the leaves. In order to identify and quantify the phenolic acids present in these extracts, chemical screening was performed by HPLC-TOF/MS, this revealed the presence of 9 phenolic acids in various amounts. The HPLC-TOF / MS analysis of the n-BuOH (leaves) extract of C. fragilis revealed the presence of 9 phenolic acids and 10 flavonoïds.
The antioxidant activity of the EtOAc and n-BuOH (leaves and flowers) extracts of C.
choulettiana was evaluated by the free radical scavenging method (DPPH) and the inhibition of lipid peroxidation (LPO). The EtOAc (leaves) extract exhibited the strongest effect with respect to the DPPH free radical (96.54%) and the inhibition of lipid peroxidation (64.17%).
Concerning the antiproliferative activity, the results showed that the cyclohexane extract of C.
choulettiana was the most active against HeLa cells (carcinoma of the human cervix), while the n-BuOH extract of C. fragilis was inactive in his unlevel doses.

Lotus roudairei specie has never been subject of a phytochemical or a biological study. The aim of the present work is isolation and structural determination of the isolated products from different extracts belonging to the mentioned species, as well as the in-vitro evaluation of the antioxidant activity and the antiproliferative activity against Human cancer cells (HeLa) of extracts from both species.
The chemical investigation of CH1, AE1 et BU1 extracts of the aerial parts of Lotus roudairei led to the isolation and characterization of 15 compounds of various skeletons: steroids, acids, flavonoids and isoflavonoids. The n-butanol extract of Genista ferox gave 3 compounds. The isolation and purification of isolated compounds were carried out by combining different chromatographic methods. Their structural determination was achieved through the use of spectroscopic techniques, such as UV spectroscopy, 1D NMR (1H, 13C), 2D NMR (COSY, HSQC and HMBC), HPLC/TOF-MS and GC/MS and by comparison with literature data.
The quantification of phenolic acids and flavonoids in extracts of both species was performed by HPLC-TOF/MS. The results revealed the presence of 13 phenolic acids and 10 flavonoids in Lotus roudairei and 44 compounds comprising 17 organic and phenolic acids and 27 flavonoids and phenolic compounds in Genista ferox.
The antioxidant activity of CH1 and AE1 extracts of Lotus roudairei was estimated by the DPPH test and β-Carotene bleaching assay. The AE1 extract exhibited the most important antioxidant activity compared to BHA used as standard. The antioxidant activity of CHT, AET, BUT and MET extracts from stems and CHF, AEF, BUF and MEF extracts from leaves of Genista ferox was evaluated by the DPPH test. The AET and AEF extracts showed significant
antioxidant activity compared with ascorbic acid used as standard.
Concerning the antiproliferative activity, the results showed that the BU1 extract of Lotus roudairei and the two extracts BUT and BUF of Genista ferox showed the lowest activity against HeLa cells (carcinoma of the human uterine cervix), while the ME1 extract of L.roudairei and the AET extract of G.ferox showed the highest activity against HeLa cells.
