Biologie Résumé : The purpose of this study is to evaluate in vivo the antioxidant and hepatoprotective effect of the butanolic extract of Lifago dielsii (BELD). The adult male Wistar Albino rats were force-fed with the BELD (250 mg / kg) for a period of 14 days and simultaneously received a combined treatment of the isoniazide (INH, 75mg / kg) and rifampicin (RIF, 150mg / kg). In vitro study of the antioxidant activity of BELD was also performed.
Combination treatment of INH / RIF induces hepatic dysfunctions that have been revealed by; a significant increase in the level of serum enzymes (ALT, AST, ALP, LDH), alteration of lipid profile parameters (HDL, LDL, cholesterol, serum TG and hepatic TG ) as well as those of hepatic nitroso-oxidative stress (MDA, 4HNE, LHP, nitrosothiols, carbonylated proteins, nitric oxide) compared to the controls. Similarly, treatment with INH / RIF induces a significant reduction in the antioxidant activity of SOD, CAT, GST, GPx and POD and the level of GSH in the liver. Histologically, the combination of INH / RIF was able to induce hepatic steatosis. In contrast, the pretreatment of rats with BELD protects the liver against the toxic effect of metabolites generated by INH / RIF, thereby maintaining liver functions and balancing hepatic redox status. It appears from this in vivo study that BELD is endowed with antioxidant and hepatoprotective activity with a positive action on the restoration of INH /
RIF-induced steatosis. The antioxidant effect of BELD has been studied in vitro by carrying out tests such as: reducing power, hydroxyl radical scavenging (HO°), total antioxidant capacity, iron chelating effect, nitrite anti-oxide effect (NO°) and Inhibition of lipid peroxidation by the ferric thiocyanate method. The results obtained show that this extract has a strong scavenger activity of radical HO° and NO°, as well as a strong reductive and inhibitory capacity of lipid peroxidation. In conclusion, the study realized valid the safety and
effectiveness of the plant lifago dielsii. Which is a plant resulting from traditional Algerian medicine. The results obtained also support the therapeutic potential of the Butanolic extract of this plant, proving that its extracts and bioactive compounds can be developed as a new natural product for the prevention of liver injuries.

Parkinson's disease (PD) is a chronic neurodegenerative disorder that develops in a slowly progressive manner with no identifiable cause. The aim of this investigation is the study of the biological potential of the n-BuOH and EtOAc fractions extracted from the plant Centaurea africana and elucidate the modulatory effect of the n-BuOH extract in rats (Wistar Albinos) treated with rotenone. In vitro studies (DPPH°, reducing power of iron, inhibition of LPO) have shown that C. africana is endowed with antioxidant properties. Anti-inflammatory studies have shown that nBuOH extract has this effect which is resulting in the inhibition of paw edema induced by carrageenan in rats and decreased activity of MPO and MDA levels in paw tissues. In vivo studies have investigated the modulatory effect of n-BuOH extract of C. africana (200 mg/kg) against biochemical, neurochemical, histopathological and behavioural alterations induced by rotenone (1 mg/kg; ip) in rats. Rotenone causes behavioral and motor deficits and also activates neuroinflammation. The mitochondrial dysfunction induced by ROT is expressed by the decrease in
the activity of complex I of the mitochondrial respiratory chain and by the alteration of the oxidant/antioxidant status of the brain, which activates the intrinsic apoptotic pathway. This study revealed that the n-BuOH extract acts by several mechanisms: neutralization of oxidative stress, modulation of markers of apoptosis (Bcl-2, Bax and caspase 3) and neurochemical markers (dopamine and glutamate), and reduction of neuroinflammation which results in decreased MPO activity and lower levels of NO and TNF. The antioxidant and anti-inflammatory properties of this
plant are probably explained by its richness in polyphenols and flavonoids, which have been revealed by phytochemical screening and LC-MS/MS analysis, and/or by the synergistic interactions of the isolated phenolics.

treated by the valproic acid (VPA). The in vitro studies (DPPH◦, FRAP, β-carotene, Chelation, NO◦) showed that Linaria tingitana is endowed with an antioxidant property. The antiinflammatory studies (albumin denaturation, HRBC, NO scavenger, oedema and peritonitis led
by the carrageenan, the rise of MPO and the infiltration of the PN) confirmed that Linaria
tingitana has a beneficial effect in this sense. The in vivo studies made to rats handled by the
VPA revealed that the n-BuOH extract acts by a multitude of mechanisms: conservation of the
hepatic function, the modulation of the steatoses, the neutralization of the oxidative stress and
the reduction of the inflammation hepatocyte elucidated by the decrease of the activity MPO.
Antirrhides molecules isolated of this plant were ineffective at the level of the cell line HepG2
treated with the VPA. The mitochondrial dysfunction led by the VPA is expressed by the decrease of the RCR accompanied by a decrease of the activity of the complex I. The n-BuOH
extract as well as the isolated molecules managed to reduce the mitochondrial H2O2 and to modulate the antioxidant mitochondrial and lysosomal systems. The conservation of the integrity
lysosomal membrane was more pronounced by the n-BuOH extract compared to the vitamin
E. The various modulator ways of L. tingitana delayed the appearance of the complications
linked to the mitochondrial dysfunction, lysosomal, such necrosis. Probably this is due to the
diversified wealth of this plant in term of polyphenols and irridoids revealed by phytochemical
screening and the analysis of the LC-MS and in the synergetic effect of the molecules that it

Biologie Résumé : The work presented in this thesis contributes to the valorization of the aerial part of an endemic plant collected from the Algerian Sahara, Heliotropium undulatum Vahl (the family Boraginaceae). This study was conducted to evaluate the antioxidant and antiinflammatory activities of n-BuOH extracts of stems and leaves of H. undulatum using a rat- hydrazine model. In vitro studies (DPPH°, Chelation, FRAP, °OH) reveal that the n-BuOH extract of leaves has an antioxidant property. In vivo studies have also shown that the n-BuOH extract of 200 mg/kg leaves has demonstrated hepatoprotective activity by preserving liver function, modulating CYP2E1, neutralizing hepatic oxidative stress, and providing intra- mitochondrial redox status exhibited during treatment with hydrazine (300 mg/kg). The antiinflammatory activity was better pronounced by the n-BuOH extract of the stems, the demonstration of which is carried out using various in vitro tests (denaturation of albumin, HRBC, NO° scavenger). The study of modulatory pathways (hepatic carbonyl proteins, MPO, levels of NO, IL-ip and TNF-a) confirmed the in vivo anti-inflammatory potential of the n- BuOH extract of stems. The set of concomitant antioxidant and anti-inflammatory mechanisms characterizing the two extracts of Heliotropiuin undulatum is correlated with the qualitative and quantitative nature of the phenolic acids and flavonoids revealed via HPLC- MASS analysis and with the synergetic effect of all the phytoconstituents contained in this plant

In vitro studies (DPPH°, Chelation, FRAP, °OH) reveal that the n-BuOH extract of leaves has an antioxidant property. In vivo studies have also shown that the n-BuOH extract of 200 mg/kg leaves has demonstrated hepatoprotective activity by preserving liver function, modulating CYP2E1, neutralizing hepatic oxidative stress, and providing intra- mitochondrial redox status exhibited during treatment with hydrazine (300 mg/kg). The antiinflammatory activity was better pronounced by the n-BuOH extract of the stems, the demonstration of which is carried out using various in vitro tests (denaturation of albumin, HRBC, NO° scavenger). The study of modulatory pathways (hepatic carbonyl proteins, MPO, levels of NO, IL-ip and TNF-a) confirmed the in vivo anti-inflammatory potential of the n- BuOH extract of stems. The set of concomitant antioxidant and anti-inflammatory mechanisms characterizing the two extracts of Heliotropiuin undulatum is correlated with the qualitative and quantitative nature of the phenolic acids and flavonoids revealed via HPLC- MASS analysis and with the synergetic effect of all the phytoconstituents contained in this plant
