Biologie Résumé : This study was conducted to search for XO inhibitors from Ajuga iva shoot extracts
(AISE) traditionally used in folk medicine in Algeria. XO was purified from fresh bovine
milk with yields of ? ? ? ? ? mg / L, protein / flavine ratio of 5.15 and a single major band, on
SDS-PAGE, of approximately 150 KDa, indicating a high purity of the purified enzyme. The
specific activity of the purified enzyme was of 2055.05 nmol / min / mg protein. The analysis
of methanol (CrE), chloroform (ChE) and ethyl acetate (EaE) extracts, showed that the EaE
has significant amount of polyphenols (35.83 ± 0.69 mg EGA / g dry extract), followed by
ChE and CrE (30.58 ± 0.2 and 17.38 ± 0.29 mg EGA / g dry extract, respectively). The ChE
has the highest flavonoids content (9.64 ± 0.33 mg EQ / g dry extract), followed by EaE and
CrE with 7.72 ± 0.19 et 4.92 ± 0.30 mg EQ / g dry extract, respectively. The ChE showed the
highest inhibitory properties on the XO activity (IC50 = 0.134 ± 0.00032 mg / ml) and on the
cytochrome c reduction (IC50 = 0.23 ± 0.0017 mg / ml), followed by EaE with IC50 of XO
inhibitory activity and an IC50 of the inhibition of cyt c reduction of 0.185 ± 0.0019 and 1.046
± 0.0055 mg /ml, respectively. Finally, CrE had the lowest inhibitory effect on XO activity
and on cyt c reduction with 0.367 ± 0.0041 and 3.34 ± 0.018 mg / ml, respectively. The
antioxidant potential was confirmed with the non enzymatic methods, DPPH assay and β-
carotene / linoleic acid bleaching assay. The highest DPPH radical scavenging activity was
observed with EaE (IC50 = 228.01 ± 2.26 µg / ml), followed by CrE and ChE with IC50 of
415.06 ± 0.59 and 466.33 ± 3.20 µg / ml, respectively. The β-carotene / linoleic acid
bleaching assay revealed that AISE have a very important antioxidant activity. Results
showed that ChE has the highest antioxidant activity (95.64 ± 0.15 %), followed by CrE and
EaE with 87.97 ± 0.10 % and 72.30 ± 0.35 %, respectively. These results suggest that these
natural products could be used to treat lot of diseases, where inhibition of XO, free radical

Biologie Résumé : The present study aimed to evaluate the protective effect of argan oil and selenium against oxidative stress induced by mercury. For this reason, a phytochemical study of antioxidant tested in vitro was performed on argan oil to estimate its expected effect. The results showed the presence of phenolic compounds in a very high concentration. The evaluation of power scavenger argan oil is more active with IC50 = 13.33 ± 1.3, and in vivo on Albino Wistar rats were treated with mercury alone or combined with argan oil and selenium, for a period of three weeks, the blood is then withdrawn and the rats are sacrificed the various parameters are determined. From the analysis of our results, we observed a significant reduction in weight gain during treatment with mercury alone or combined with argan oil and selenium, but no clear change in the relative weight of liver, kidney or spleen was obtained. The results also showed significant changes in biochemical parameters, immunological and haematological. Characterized primarily by a significant increase in serum glucose, urea, creatinine, uric acid, total and direct bilirubin, total lipids, cholesterol, triglyceride, conjugated diene and the enzymatic activity of AST, ALT, LDH and PAL .dropoff window Whereas serum concentrations of albumin and total protein were reduced in treated rats compared to mercury control animals. Results also show immunological and haematological toxicity especially among the treated groups the pure mercury, an increase in serum IL-1, IL-6 and TNF and elevated white blood cell count, decreased red blood cell levels, hemoglobin concentration the percentage of hematocrit and phagocytic activity during treatment with mercury. Thus the treatment of the rats with the mercury causes a reduction in the plasma antioxidant capacity characterized by decrease in the serum concentration of vitamin E and C and rate of DPPH and thiols. We also observed that the results obtained highlight the detoxifier potential expressed by the decrease in erythrocyte glutathione levels, liver and kidney, metallothionein, the enzymatic activity of the antioxidant system SOD, GPx, GST and catalase and increased in lipid peroxidation expressed by the high levels of plasma MDA, erythrocyte, liver and kidney. Also the results clearly showed substantive alterations of tissue studies in comparison with the control, with hepatocyte cytolysis, hepatic necrosis and tubular degeneration with necrosis of the kidneys. Furthermore supplementation argan oil and selenium changed the toxic effects of mercury and decreased histological alterations and showed that both antioxidants are effective against oxidative stress induced by mercury

Aerial part of Pituranthos chloranthus showed a moisture of (11,8 ± 0,097) % and an essential oil’s extraction yield, extracted by hydrodistillation, is near (0,86 ± 0,09) % and an optimization of time (t = 250min) for a dry plant mass of de (m = 80 g).
Antioxidant activity of the present plant material, determined by three tests: reducing power,DPPH and
-carotene bleaching test against a standard (α-Tocopherol or Vitamine E). The reducing power is of (0.094 ± 0.018) g TE/g EO, IC50 of (0,024 ± 0,002) mg/ml and a protective effect against
-carotene oxidation.
Physico-chemical characteristics of the shortening produced including: acidity, peroxide index and melting point are in a good agreement with the internal company standards.
Assessment of oxidative stability is carried out by Rancimat and Schaal tests. Rancimat showed that shortening incorporated with 75ppm gave the best resistance to oxidation. This is also consistent with Shaal test, with a peroxide index of PI = 0, 28 meq g O2/1000g, carried out in 35 days.
Thus, Pituranthos chloranthus essential oil may be of a technological interest to serve like preservatives in fats and oils, especially in food companies.
